电针结合康复训练在周围神经损伤后功能重建的临床康复中应用广泛,然其作用机制尚不明确。Rho族GTP酶(Rho GTPases)是轴突导向信号通路上的关键调节分子,能通过诱导细胞骨架重组影响轴突再生。近年研究证实:Rho GTPase 的失衡受排斥及吸引性导向因子的调控。我们提出:“电针结合神经松动术能调控周围神经损伤后Netrin-1和Slit2共同表达,调整了失衡的Rho GTPase,促进轴突再生”之假说。本项目建立兔坐骨神经损伤模型,从形态学、神经电生理、分子生物学等角度,研究电针结合神经松动术促进轴突再生与Rho GTPases失衡的关系;构建netrin-1和Slit2病毒,研究netrin-1、Slit2共同表达与Rho GTPases失衡的关系。探索电针结合神经松动术促进周围神经损伤后轴突再生的机制,为其临床应用提供理论基础。

Electro-acupuncture combines with rehabilitation medicine is applied wildly to treat peripheral nerve injury in clinical care, but its mechanism is still unclear. The Rho GTPases is a key regulating molecules to effect on axon regeneration via inducing reorganization of cytoskeletion. Recent studies suggest that the unbalance of Rho GTPases is regulated by signal of repulsion and attractiveness. We hypotheses that electro-acupuncture combines with neurodynamic mobilization may regulate the co-expression of Netrin-1and Slit2, also adjust the unbalance of Rho GTPases to facilitate axon regeneration. We will make the models of sciatic nerve injury of rabbits to research the relationship between axon regeneration and unbalance of GTPases, also the relationship between co-expression of Netrin-1and Slit2 and unbalance of GTPases by using electro-acupuncture combines with neurodynamic mobilization. Exploring the mechanism of treatment of electro-acupuncture combined with neurodynamic mobilization on axon regeneration after peripheral nerveinjury in order to provide a theoretical basis for its clinical application.
