胰腺癌预后极差,是最难治的恶性肿瘤之一。申请人针对胰腺癌乏氧微环境的病生理特征,开展系列基础与转化研究,重点探讨HIF-1参与胰腺癌恶性生物学行为的分子机制,结合纳米技术探索控制策略,旨在突破胰腺癌临床诊治瓶颈。近五年主要贡献:1)揭示了HIF-1参与胰腺癌增殖、转移、代谢的分子机制,探讨HIF-1抑制剂联合化疗的分子机制;2)设计新型纳米载体药物,提高胰腺癌化疗疗效,具有重要临床转化前景;3)结合临床病理特征,探索胰腺癌诊断及预后的分子标志;4)立足临床外科,针对胰腺癌不同治疗策略进行研究,为进一步规范胰腺癌临床诊疗,制定中国版胰腺癌诊疗指南提供依据。近五年在Advanced Functional Materials, Cancer Research, Biomaterials 等杂志发表SCI文章28篇,其中通讯作者和第一作者(含共同)文章26篇,获聘天津市特聘教授,教育部新世纪人才。
英文摘要
Pancreatic cancer is one of the most lethal human malignancies with extremely poor prognosis. Based on the characteristic hypoxia microenvironment, the applicant developed a series of basic and translation researches. He focused on the identification and validation of Hypoxia Inducible Factor-1 (HIF-1)’s function in progression of pancreatic cancer, and searching for the corresponding strategy by using nanotechnology, thus to break through the bottleneck of its diagnosis and treatment. Major contributions of the PI in the last five years are as follows: 1, He elucidated the molecular mechanism that HIF-1 facilitates proliferation, metastasis and metabolism of pancreatic cancer. Meanwhile, he firstly reported the the mechanism of HIF-1 inhibitor based combinational chemotherapy. 2, By encapsulating both lipid-soluble and water-soluble particles, he developed new chemotherapy drugs with nanocarrier, which showed great translational potential for the improvement of chemotherapy. 3, Combined with the characteristics of clinical pathology, he identified several new biomarkers of the diagnosis and prognosis of pancreatic cancer. 4, He developed different therapeutic strategies for pancreatic cancer which are used in the patient care in his department. Moreover, in order to draw up the Chinese pancreatic cancer treatment guidelines, he made further standardization to the clinical pathway of pancreatic cancer. For the recent five years, the applicant has published 28 articles in Advanced Functional Materials, Cancer Research, Biomaterials, etc. 26 of these were published as corresponding or first author (Co-author included). He was selected as Distinguished Professor in TIANJIN and Talents of the New Century by the Ministry of Education.
