肿瘤转移是影响肿瘤患者预后及治疗的关键因素。申请人以个性化的结直肠癌转移动物模型出发,从结直肠癌侵袭转移的分子特征着手,结合临床验证和功能研究,在结直肠癌转移相关分子特征和机制等方面进行了系列研究:(1)建立基于肿瘤细胞亚克隆不同生物学特性筛选结直肠癌转移相关分子特征的研究策略;发现SATB2和一个分子标签在结直肠癌转移中的作用。(2)发现PRL-3在结直肠癌中通过对钙粘附蛋白的直接作用参与EMT 的新机制、与微管相关蛋白Stathmin之间的功能联系;揭示PRL-3调控结肠癌细胞表观遗传特性的机制。(3)发现Nanog等“干性”分子在结直肠癌中的作用。(4)发现EGFR、TGF-beta等的调控新机制、新效应;发现Erbin 在结直肠癌发生及早期侵袭的作用。近5年以通讯作者发表SCI论文12(IF63.8)。

Metastasis is one of the key factors affecting the prognosis and treatment of cancer patients. We have made a series of works on the screening and functional studies of metastasis associated molecules of colorectal cancer (CRC): (1) We have well established the strategies for screening and functional validation of metastasis a ssociated molecules in CRC based on phenotypes in different cell subclones. The roles of SATB2 and a molecular signature in CRC have been discovered and validated. (J Pathol, 2010 et al.) (2) As an important biomarker for metastasis and prognosis of cancer patients, several mechanisms of PRL-3 involved in CRC was found. Interestingly, recently we uncovered an epigenetic role for PRL-3 as a regulator of Histone H3 methylation. (Gut, 2013; J Proteome Res, 2010 et al.) (3) A key role, clinical significance, reprogramming potential of Nanog was found in CRC (Cancer Biol Ther,2010 et al.). (4)The new mechanism or effects of EGFR or TGF-beta signaling were found. Erbin was first suggested as a key player in early invasion and pathogenesis of CRC (J Pathol, 2014; Oncogene 2014 et al.) . The applicant has published 12 peer-reviewed papers (Impact factors 63.8) as the corresponding author in the past five years.
