慢性气道炎性疾病患者的气道黏液呈爆发性高分泌状态,而此爆发性分泌与粘蛋白(主要为粘蛋白5AC)的出胞运动增加有关。Exocyst复合物则是介导分泌囊泡与胞膜栓系的关键拴链因子,因此,研究粘蛋白的分泌出胞运动,Exocyst复合物是最佳候选靶目标。冷空气刺激是COPD患者急性加重的重要诱导因素之一,而冷刺激引起粘蛋白出胞运动增加导致气道黏液分泌增多的作用机制尚未被研究。本交流合作项目以面上项目“拴链因子Exocyst复合物在慢性气道炎症中的促黏液分泌效应”研究为依托,借助该项目对Exocyst复合物在气道上皮的表达、定位及在粘蛋白分泌出胞中的作用研究基础,利用在研项目的既有成熟的体外细胞培养技术,将进一步完善寒冷刺激对气道上皮黏液高分泌效应的信号分子,揭示Exocyst复合物在粘蛋白出胞分泌过程中的作用机制。以拴链因子Exocyst复合物作为靶点并利用俄方的寒冷气候作为研究的有利条件,将我方的黏液分泌机制研究与俄方的冷空气刺激相关研究有机结合起来,进行协作研究。此合作课题把双方的研究优势巧妙结合,有利于各自在研项目的深入开展,同时课题的新颖性也有助于增强双方在相关领域的研究实力及影响力。

Airway mucus was in explosive high secretion state in the patients with Chronic airway inflammation, and this explosive secretion is related to mucin (mainly mucin 5AC) increased motility of the cell.Exocyst complexes are the key binding factors that mediate the secretion of vesicles and membrane tethers. Therefore, to study the mucin secretion cell motility,Exocyst complex is the best candidate target.Cold air stimulation is one of the important inducing factors of acute exacerbation of COPD patients, but the mechanism of increased airway mucus secretion induced by cold stimulation of mucin motility has not been studied.This project is based on the research project of the "Exocysts in Chronic Airway Inflammation to Promote Mucous Secretion".With the help of the project on the Exocyst complex in the airway epithelial expression, localization and secretion of mucin secreting cells in the role of the foundation.Using the mature cell culture technique in vitro, the signaling molecules of airway epithelium mucus hypersecretion can be further improved by cold stimulation, and the mechanism of Exocyst complexes in mucin secretion can be revealed.Using the tethered factor Exocyst complex as a target and the cold climate of Russia as a favorable condition, to combine with the Study on the Secretory Mechanism of us and the Research on Cold Air Stimulation of Russia,to collaborate.This collaborative project cleverly combines the advantages of the two sides of research,which is conducive to their in-depth research projects carried out.At the same time the novelty of the subject will also help to strengthen the research strength and influence in the related fields.
