本项目是在对杜松种群生态学系统研究的基础上,针对杜松种质资源保护与利用的迫切需求,以及种质资源研究中的关键或难点问题入手,利用形态标记、分子标记(SSR和SRAP)、化学指纹图谱标记(HPLC)、GIS等技术,运用历史资料的整合、详尽的野外调查取样、“同质园实验”和化学计量学等多种方法,以及相关学科的理论与方法,从表型、分子、生理(化学)3个水平探讨杜松种群遗传结构和遗传多样性;检测杜松不同部位化学成分的差异和不同产地主要药用部位主要化学成分含量的变化;以抗氧化、抗真菌为考察指标,研究杜松生物活性的地理变异;结合生物数据和地理数据,应用GIS和Fuzzy构建的评价模型,对杜松种质资源进行适宜性评价。本研究旨在建立杜松种质资源数据库和综合评价体系、阐明杜松居群遗传变异特征及规律、建立杜松种质资源区划主题图、筛选出优良种源地、制定保护与保存策略,为将来的高效利用提供基础。

In this project, based on the systematic research of Juniperus rigida population ecology, according to the urgent requirements of protection and utilization of Juniperus rigida germplasm resources, starting with the key and difficult problems in the study of germplasm resources, using morphological markers, molecular markers(SSR and SRAP), chemical fingerprint markers (HPLC) and GIS analysis, using the integration of historical data, detailed field investigation and sampling, “common garden experiment”, chemometrics methods and related theories, Juniperus rigida population genetic structure and genetic diversity from phenotypic, molecular and physiology (chemistry) three levels are explored; the differences of chemical composition of the different parts and the changes of main chemical composition content from different main medicinal parts and regions are detected; with the antioxidation and antifungal effect as indexes, geographic variation of Juniperus rigida biological activity are studied; combined with biological data and geographic data, application of GIS and Fuzzy construction of model, the habitat suitability of Juniperus rigida germplasm resources are evaluated. This study aims to establish a database of Juniperus rigida germplasm resources and comprehensive evaluation system, explain characteristics and regularities of Juniperus rigida population genetic variation, establish a topic map of Juniperus rigida germplasm resources division, screening excellent provenance, formulate protection and preservation strategy, thereby making a basement to efficiently utilization in the future.
