申请者从事中药抗肿瘤和防治神经退行性疾病药效物质基础的研究。基于活性评价及化学筛选集成策略,对蜘蛛香、天麻等8种中药的药效物质进行再研究:获得了不同结构类型的新中药化学成分242个,4个为新颖骨架化合物,其中2个被国际权威综述刊物Nat. Prod. Rep. 遴选为热点道;发现具有显著抗肿瘤和神经保护等活性成分30个。在此基础上,首次揭示了蜘蛛香“活血消肿”、天麻“益智”的药效物质基础;并从蜘蛛香中获得了一类高效、低毒抗卵巢癌的环烯醚萜类活性物质,阐明了其作用机制。在Org. Lett., Chem. Commun., J. Nat. Prod.等本领域顶级期刊发表SCI论文45篇,包括第一/共同通讯作者论文16篇,2篇影响因子大于6,单篇他引最高次数为30。入选“北京市优秀人才”和北京协和医学院“协和新星”。拟继续进行防治神经退行性疾病的中药药效物质基础及其作用机制的系统研究。

The applicant was engaged in therapeutic basis of traditional Chinese medicines. 242 new compounds have been isolated and identified from eight traditional Chinese medicines by application of bioactivity-directed fractionation combined with HPLC-PDA-MSn and HPLC-NMR online detection. Four novel compounds with unprecedented skeleton also have been found, of which fraxinuacidoside and yaoshanenolide A were selected as the hot of the press in the review of Natural Products Reports. Most of the isolates were subjected to bioactivity evaluation in different models, leading to the discovery of 30 novel compounds with remarkable anti-tumor, anti-inflammatory, and neuroprotective activities. One of the iridoids, IVHD-valtrate, isolated from Valeriana jatamansi, inhibited the growth of A2780 and OVCAR-3 xenograft tumors in nude mice with minimal toxicity. The in-vitro and in-vivo mechanism studies indicated that IVHD-valtrate activated the p53 signaling pathway and regulated genes associated with cell growth and proliferation, apoptosis and cell cycle progression. A series of bibenzyl compounds with neuroprotective activities were found in Gastrodia elata for the first time. In all, 45 papers in chemistry and pharmacology have been published, including 16 with first author or co-corresponding author in Organic Letters, Chemical Communications, etc. The applicant was selected as the "Beijing outstanding talents cultivation fund" and "Peking Union Medical College nova program" in 2013. The future research will still focus on the therapeutic basis of TCM for neurodegenerative diseases.
