促进缺血性卒中的康复有重大的社会和经济意义,探寻其神经修复调控的新途径是目前关注的重点。当今热点领域表观遗传学最新研究表明:由脑缺血诱导的MeCP2磷酸化修饰及以MeCP2为中心的信号途径在神经干细胞自我更新与分化、神经元分化成熟与基因调控等方面扮演重要角色。课题组前期研究证实针刺能激活脑缺血组织NGF/VEGF/TGFβ等基因表达而改善大鼠神经功能。本项目围绕脑缺血损伤神经修复这一环节,以MCAO大鼠为观察对象,以电针心经心包经为干预手段,紧扣以MeCP2为中心靶标的在神经干细胞分化调节中的两个重要信号途径(MeCP2/miR-137、MeCP2/BDNF),利用前沿的RNA-seq、ChlP-seq及其它分子生物学、神经生物学技术,从NDA甲基化修饰途径探索针刺心经心包经在脑损伤过程中参与神经修复效应的调控机制,为"心-心相关经脉-脑关系"这一中医原创理论的科学内涵提供全新阐释。

Promoting the recovery of Ischemic stroke is of great significance to the society and economy and exploring the new way of nerve repair regulation is still the focus of attention. Researches in epigenetics, today's hot field, show that the MeCP2 phosphorylation induced by cerebral ischemia and MeCP2-centered signaling pathways plays an important role in the self-renewal and differentiation of neural stem cell, in the maturity of neuronal differentiation and gene regulation. Early studies in our group have confirmed that acupuncture could improve the neural function in rats by activating the gene (NGF,VEGF,TGFβ,etc.) expression of the cerebral ischemic tissue. This project focuses on repairing the damaged nerve from cerebral ischemia, by needling the heart and pericardium meridians of MCAO rats for intervention, targeting MeCP2's two important signaling pathways (MeCP2 / miR - 137, MeCP2 / BDNF) in the regulation of neural stem cells differentiation, exploring the regulatory mechanism of nerve repair effect in the brain injury from DNA methylation modification with the help of a series of advanced technology such as RNA - seq, ChlP - seq and others related to molecular biology and neurobiology to provide new interpretations for this original theory of TCM "relationship among the heart,heart - related meridians and the brain".
