异皮亚科(Heteroderinae)线虫在多个国家发生并造成巨大经济损失,全世界已报道此类线虫8属116种,我国仅报道3属18种。基于我国地域辽阔性和物种资源多样性,本研究将采用形态学、数值分类方法和分子系统学技术,对我国异皮亚科线虫进行系统的分类研究,同时将收集到的种群进行rDNA、mtDNA和微卫星DNA等序列分析,结合Genbank中相关数据构建该亚科下属、种的系统进化关系。利用GIS技术,制作异皮亚科中以大豆胞囊线虫为代表类群的不同种群基因型归类和地理分布图,以分析我国大豆胞囊线虫地理分布的时间动态、扩散和传播规律。本课题研究结果将进一步丰富我国异皮亚科线虫种类资源,明确该亚科线虫种群和系统进化关系,探索异皮亚科中重要病原线虫-大豆胞囊线虫的地理分布特征和传播规律,为异皮亚科中重要病原线虫在农业生产中造成病害的防控提供基础研究数据。
英文摘要
Some species of Heteroderinae can make heavy economic losses in many countries around the world, there were reported 8 genera and 116 species worldwide, however, in China only reported 3 genera and 18 species because there are only few scientist focus on nematode taxonomy. The project will integrate classic morphological and molecular techniques for finding new species of Heteroderinae all over China. The new species and new record species will be identified by morphology, measurement, microscopy and scanning electron microscopy,the speciman will be extracted rDNA, mtDNA and Microsatellite DNA, then amplification and sequencing, combined with the the Genbank existing data to construct phylogenetic relationships of the different genera and species. Distribution map of soybean cyst nematode as a representative of Heteroderinae will be drawn by GIS. Geographic distribution, diffusion and spread will be analysis according to integrating geographic and genotype data of H. glycines. This project will further clarify numbers of cyst nematode species and phylogenetic relationships, and provide study foundation for management these pests.
