杆状病毒被广泛应用于生物农药、蛋白质表达以及疫苗生产。杆状病毒的多个蛋白在细胞质翻译后转运到细胞核,参与病毒的复制和装配,但是,这些蛋白从细胞质进入到细胞核这一过程仍不清楚。本申请项目以杆状病毒的DNA聚合酶为研究对象,通过荧光免疫、肽融合、位点缺失和突变、酵母双杂交等实验方法,开展以下方面的研究:(1) 杆状病毒DNA聚合酶核定位信号的研究;(2)Sf-9细胞输入蛋白的克隆;(3)DNA聚合酶核定位信号与Sf-9细胞输入蛋白相互作用的鉴定。这些研究将对理解杆状病毒蛋白的入核机理具有重要意义。

Baculovirus were widely used in biopesticide, protein expression and vaccine production. Most of baculovirus proteins were involved in baculovirus DNA replication and assembly, these proteins express in cytoplasm and are transported into nucleus. However, it is unclear how the proteins were imported into nucleus. In this work we will focus on the several issues using immunofluorescence, peptide fusion, multiple point substitution, deletion mutation and yeast two-hybrid system :(1) nuclear localization signals of baculoviru DNA polymerase; (2) the clones of importin in Sf-9 cells; (3) the interaction between nuclear localization signal and importin. The results will contribute to the understanding of nuclear transport of baculovirus proteins.
