腰椎间盘突出症是基层医疗卫生服务机构的重要疾病负担,前期研究发现,采用疏肝补肾法研制的腰腿痛胶囊疗效显著,但作用机制不明。p38 MAPK在介导细胞炎性介质的释放中发挥重要作用,其变化影响“疼痛信号”的传递。NF-κB作为p38MAPK下游炎性信号相关的细胞转录因子在炎症反应中起重要作用。本研究建立“髓核渗漏刺激DRG造成的腰突症的疼痛,可能是通过P38MAPK/NF-κB信号转导通路的交互响应导致DRG疼痛敏化引起”的工作假说,拟以自体髓核移植大鼠模型作为研究对象,采用Elisa,EMSA,Western-blot,RT-PCR检测DRG及血清中不同时间节点相关因子的表达,探索DRG中P38MAPK/NF-κB信号转导通路关键分子的表达与IL-lβ、IL-6、TNF-α、SP、CGRP之间的关系,揭示腰突症疼痛的发生机制及疏肝补肾法对其干预的作用。

Lumbar disc herniation is an important disease burden of basic medical and health services institutions ,Based on the previous research, the curative effect of Yaotuitong Capsule developed through Shugan Bushen method is significantly, but the mechanism is unknown.but the mechanism is unknow.p38 MAPK plays an important role in modulation of inflammatary reaction. Active p38 can induce NF-κB translocation, which promted mutiple genes transcription.The impact of the change transfer "pain signals". NF-κB as p38MAPK Preparation of the inflammatory cells associated transcription factors play an important role in inflammation. This study established "nucleus stimulation through interactive response p38MAPK/NF-κB leakage signal transduction pathway may be one reason for the occurrence of injuries sensitized DRG pain lumbar herniation pain" working hypothesis,proposed autologous nucleus pulposus rat model of transplantation as the research object, using Elisa EMSA Western-blot RT-PCR to detect DRG expression, and serum nodes in different time-related factors, exploring the relationship between p38MAPK/NF-κB key signal transduction pathways and expression of molecules IL-lβ,IL-6,TNF-α,SP,CGRP in DRG, reveals the pain mechanism in traumatic lumbar disc herniation of shu gan bu shen fa and its method of intervention.
