最新研究发现,长链非编码RNA(LncRNA)在部分疾病中发生了异常,这些异常的表达或调控对疾病有重要的影响。但LncRNA与大部分疾病的发生、发展的关系及作用机制未被阐明,包括LncRNA与骨质疏松之间的关系。线粒体是细胞生命活动的控制中心,也是细胞凋亡的控制中心。我们前期研究发现骨骼肌中线粒体和骨质疏松之间存在着互相影响的关系。本研究①拟采用LncRNA-m RNA 基因芯片技术对正常及去势大鼠骨骼肌线粒体内的 LncRNA 表达情况进行研究,获得其特异表达谱,并进行调控网络分析。③采用基因超表达和沉默技术及相关分子生物学方法,研究中药靶向目标LncRNA 在调控成骨细胞增殖防治骨质疏松症中的作用。探明目标LncRNA在人、动物及细胞中的表达情况,以及与骨质疏松的关联性,为后续骨质疏松症相关 LncRNA 的研究提供数据基础和方向指导。

The latest study found that some abnormal expressions occur in Long non-coding RNA (LncRNA) in some diseases, these abnormal expressions and regulations have a fatal influence on such diseases. However,the relationship between the LncRNA and the mechanism of those diseases has not been elucidated,especially in osteoporosis. Mitochondria is the control center of cell activity,and apoptosis. Our previous study found the interplay of osteoporosis and mitochondria in skeletal muscle..This study will firstly adopt LncRNA-m RNA gene chip technology to investigate the expression of LncRNA in mitochondrial in skeletal muscle both in normal and ovariectomized rats,to acquire the specific expression profile and analysis the regulation network,secondly,adopt gene overexpression and silencing technology or related molecular biological methods to investigate how our Chinese herbal compound impact target LncRNA in regulation of preventing and treating osteoporosis by osteoblast,finally to explore the expression of the target LncRNA in human, animal and cell, as well as the connection with osteoporosis, to provide data basis and guidance for the follow-up study.
