从"肾主骨生髓"的中医经典理论出发,基于补肾方可通过改善骨质疏松动物骨折后MSCs动员而促进骨愈合的前期研究结果,再结合"SDF-1/CXCR4 轴依赖的MSCs定向迁移是骨损伤修复的重要环节"的进展,提出补肾法可通过SDF-1/CXCR4轴介导的干细胞募集来促进骨质疏松性骨折愈合的假说。利用细胞培养技术,BrdU细胞标记示踪技术与显微CT技术等,通过体外、体内实验,观察补肾方及其温肾阳/养肾阴拆方对骨质疏松性骨折愈合的作用,以及对于SDF-1/CXCR4轴相关因子表达、MSCs定向迁移的影响;观察骨质疏松性骨折动物是否存在SDF-1/CXCR4轴介导干细胞募集行为改变进而影响骨损伤修复机制;明确补肾方是否通过SDF-1/CXCR4轴来加强干细胞的骨损伤修复机制;进而探索温肾阳/养肾阴组分对于前述过程的影响,为丰富骨质疏松性骨折治疗的中医理论和充实"肾主骨生髓"的现代科学内涵提供实验依据。

Based on the theory of “kidney storing essence”, the effectiveness of Tonifying-kidney Formula in the treatment of osteoporosis and the results of clinic study on osteoporotic fractures, and mobilization and migration of mesenymal stem cells (MSCs) are mainly controlled by stromalcell-derived factor 1 (SDF-1) and its receptor CXC chemokine receptor 4 (CXCR4). We put forward a hypothesis that the effectiveness of TKF for osteoporotic fractures is based on the migration of mesenchymal stem cells regulated SDF-1/CXCR4 axis. So, the proposal to research the effect of TKF on callus formation, and behaviors of MSCs in the process of healing of osteoporotic fractures. The aim of the study is looking for proof to support our hypothesis and questing further evidence for the “kidney storing essence” theory, and enriching the knowledge of the treatment of osteoporotic fractures.
