伸筋易骨法是中医骨伤筋骨并重思想在临床的具体应用,是膝关节骨性关节炎(KOA)有效的治疗手段,前期临床研究表明伸筋易骨法可明显缓解KOA患者临床症状。前期实验研究表明伸筋易骨法可调节膝关节周围软组织的应力平衡,减少软骨细胞凋亡。本研究基于软骨细胞力学-生物信号转导通路,采用全细胞膜片钳、免疫组化、RT-PCR等技术观察伸筋易骨法对兔KOA软骨细胞膜钙离子通道、细胞内钙离子浓度、甲状旁腺激素相关蛋白(PTHrP)及其转录基因表达的影响。揭示伸筋易骨法影响软骨细胞代谢、防治KOA的内涵,论证中医筋骨整体调节的科学意义,探究应力影响软骨细胞的生长、增殖和代谢的机制,为伸筋易骨法的科学应用提供依据。

Shenjinyigu is the Chinese medicine though that pays equal attention to bone and muscle,which is an effective treatment for the knee osseous arthritis (KOA), Clinical studies show that Shenjinyigu can significantly alleviate the patients' clinical symptoms.Experimental studies indicated Shenjinyigu improves function of unbending, adjusts knee soft tissue stress balance, protects articular cartilage, reducing the chondrocyte apoptosis. This study uses the entire membrane patch clamp, Immunohistochemical,RT-PCR to observe the impact of Shenjinyigu manipulation on KOA in rabbit cartilage cell membrane calcium channels, Intracellular concentration of calcium ion,PTHrP and PTHrPmRNA. To annonce how the Shenjinyigu To affect the metabolism of cartilage cells,controlling KOA. This provided scientific basis for clinical usage of Shenjinyigu.
