群体坚持以“正常黏膜—癌前损害—口腔鳞癌—整复再生”临床疾病自然衍进与诊治序列中多分子事件及其临床意义为核心,在癌变转移新标志、靶向治疗与干预和干细胞分子调控等方面取得原创性成绩。建设期内,发表SCI论文247篇,包括领域权威期刊,也包括生物医学领域的知名期刊Nat Commun、ACS Nano等;新增优青1人、万人计划2人、创新领军人才1人、百千万人才1人、省级青年团队2个,引进青千2人;共同创办的高水平全英文学术期刊IJOS和BR,双双位列领域内亚洲第一。总之,群体在科学研究、团队与学科建设、国际合作与交流等方面工作状态良好,具备了进一步跃升的能力;若获连续资助,群体拟深化交叉合作,从细胞模型、动物模型、结合临床队列深化研究,力争在癌变转移新分子的功能、靶向干预潜能、再生调控新分子等三个方面,取得新突破,为实现口腔癌前损害个体化精准治疗、“全方位全周期保障人民健康”,贡献力量。

英文摘要

Over the past 3 years, our team has strived to study the molecular biomarkers in the different stages of progression and recovery from oral premalignant lesions to squamous cell carcinoma (OSCC), including normal mocusa, oral premalignant lesions (OPL), OSCC, and recovery after intervention, especially focusing on the novel multimolecular and cellular events with high clinical significance. We identified several new biomarkers for metastasis of OSCC, such as PA28γ、ORAOV1、RACK1, which also served as excellent predictor for poor clinical outcomes. Mechanistically, we found that newly identified variants of PA28γ and ORAOV1 were associated the poor prognosis in the patients with OSCC. As well, the upregulation of RACK1 induced by EGFR signaling contributed to the locally uncontrollable inflammation at sites of OSCC. Regarding to the targeting therapy, we developed novel nucleosides targeting viruses associated with OSCC. By applying nano-drug delivery system, we further showed that the nucleoside targeting PA28γ suppressed CCL2 and IL-6. As of teeth regeneration, we found GDF11 inhibited the osteogenic differentiation of MSC by TGF-β signaling, and DNA methylation was involved in the tooth development..During this period, we were approved for National Clinical Research Center for Stomatology, Innovative Research Team of Ministry of Science and Technology of China, National Key Clinical Specialist Department of China, and 111 Project of Ministry of Education of China, and for 20 more research projects (about 28 millions). Our team published 247 Science Citation Index (SCI) papers, in total, on not only leading journal in our field, but also other top journals, such as Nat Commun, ACS Nano, in which 63 papers were cooperated by at least two members of this project. We also published 4 academic monographs, including 1 English one. Besides the acquired 14 patents, our team is now applying for another international patent. Our achievements set 9 new standards and an international instruction for professionals. And our team was awarded 1 national academic award for the second class and 3 awards of the first class at state level..In addition to team awards, individual members have already achieved significant progress. For instance, 1 member was awarded by Science Foundation for The Excellent Youth Scholars of Ministry of Education of China, and 2 were awarded for ten 1000 Talents Plan. 1 member was awarded for leading scientists, and 1 was awarded for BaiQianWan Talents Program. 2 subgroups were awarded for Youth leading research team of Sichuan province. To build up a world leading team, we also adopted “Recruitment Program for Young Professionals” for 2 researchers. Moreover, we published 2 English scientific journals, IJOS and BR, which are ranked 1st in Asia. As of international cooperation, we hosted international conference for 8 times, and published many papers with collaborators in the first class journals, including Cancer Cell (cover story), Nature Genet, Nat Med, Nat Immunol, Nat Cell Biol, Nat Commun, JNCI, JBMR and Cancer Res etc.
