小麦禾谷孢囊线虫是小麦生产上的重要线虫病害,严重威胁着我国的粮食安全。植物线虫分泌的钙网蛋白(Calreticulin)在致病和抑制寄主免疫过程中起着关键作用,但作用机理和调控机制尚未明确。在前期研究基础上,通过HIGS和过表达技术解析禾谷孢囊线虫钙网蛋白Ha-CRT1在寄生和致病中的关键作用,采用免疫组织定位和亚细胞定位技术研究Ha-CRT1在寄主内的作用位点,利用酵母突变体系研究Ha-CRT1抑制Bax引起的细胞死亡所依赖的信号通路,采用免疫共沉淀结合质谱分析技术鉴定Ha-CRT1的互作蛋白,采用BiFC和GST Pull Down进行互作验证。从而明确Ha-CRT1在寄生和致病中的生物学功能和调控寄主先天免疫反应的分子机制。其研究结果将探究植物线虫致病分子机理和调控寄主PTI抗性的作用机制提供新的证据,为制定科学、合理的防控策略和保障我国粮食安全具有十分重要的理论价值和实用意义。

The cereal cyst nematode (Heterodera avenae) is an important plant parasitic nematode disease on wheat, it caused serious yield losses of wheat and potential threaten to wheat production and food security in China. The Calreticulin(CRT)secreted by plant parasitic nematodes have been shown to play a role in the pathogenicity and modulation of host defenses but the mechanism of action and regulation is not clear. In this study, we investigate the function of Ha-CRT1 during the plant parasitism and pathogenicity using HIGS and over-expression. The action site of Ha-CRT1 in host cell will be tested by immunodetection and subcellular localization, research which signaling Ha-CRT1 inhabit Bax induce cell death depend on in yeast, identify the physically interact proteins of Ha-CRT1 by co-immunoprecipitation and LC-MS/MS assay, confirm the interaction between host interact proteins and Ha-CRT1 by BIFC and GST Pull Down and determine the molecular mechanism of Ha-CRT regulate host innate immunity via RNAi. The results will supply evidences to understand the interaction molecular mechanism between cereal cyst nematodes and host, supply the new evidences for the regulatory mechanisms of the host immune response by plant nematode effector and it is very important beneficial to make decision control strategies.
