The therapy of peripheral nerve defect have always troubled neuroscience and orthopedics.It is very important for the peripheral nerve defects repair that establish the allogenic nerve tissue bank, and that the cryopreservation of peripheral nerve is the key to establish the allogenic nerve tissue bank. Cryopreservation can lead to tissue and cell damage. In our previous studies (NSFC:81373668), Ligustrazine, ginseng saponin Rb1 and tripterygium glycoside can inhibit the apoptosis of peripheral nerve SCs and increase the activity of the peripheral nerve in cryopreservation, but the defect compared with the fresh nerve is very serious. The toxic effects of CPA and the low temperature and hypoxia, as well as the anti vitrification and Dilution infiltration, may cause severe damage to the peripheral nerves in cryopreservation, and preventing the occurrence of these injuries may be the key to the cryopreservation of peripheral nerves.. Cold induced RNA-binding protein (CIRP) is a kind of cold stress protein, which has a protective effect for the body in low temperature, hypoxia and osmotic stress. Ligustrazine and panaxosides can increase tissue ischemia and hypoxia tolerance. The central theme of this project proposal is to prevent the damage of the peripheral nerve by the expression of CIRP in cold preserved peripheral nerve and the effect of ligustrazine, ginseng saponin Rb1 and tripterygium glycosides, and to repair the injury of the peripheral nerve in vitro before nerve allogeneic transplantation.
