诱导膜技术是治疗各种原因引起的骨缺损的有效治疗方式。如何促进成骨及矿化,加速诱导膜形成是亟待解决的问题。近年来,对内源性骨生长因子在诱导膜形成过程中的表达和意义,以及通过导入外源性骨生长因子诱导膜形成成为研究热点,虽有一定效果,但临床应用尚有较大差距。成骨细胞是骨形成的主要效应细胞,促进成骨细胞分化及矿化将有助于提高成骨效应。本课题拟建立诱导膜动物模型,从Wnt/β-catenin信号通路的调控来研究骨碎补总黄酮促进诱导膜形成的微观机制。从形态学和分子生物学角度阐明骨碎补总黄酮对诱导膜形成和Wnt/β-catenin信号通路的影响,丰富了中医药补肾壮骨的理论,也有利于客观评价中医药在促进成骨方面的作用。

Induced membrane technique is currently an effective therapeutic option for osteomyelitis, osteosarcoma, and various congenital or acquired bone deformities.How to promote bone formation and mineralization to accelerate induced membrane formation is a serious issue yet to be solved. In recent years, the significance of endogenous expression of bone growth factor in the process of induced membrane and exogenous introduction of bone growth factor to promote new bone formation has been intensively studied. Despite of progress made in this research field, the clinical application is still challenging. Osteoblast is major effector cell of distraction stess, which can promote osteoblast differentiation and mineralization to improve osteoblasts. This project is proposed to establish an osteoblast distraction model to investigate the molecular mechanism of osteopractic total flavone in promoting induced membrane formation through Wnt/β-catenin signaling pathway. Furthermore, elucidating the role of osteopractic total flavone in osteoblast proliferation/differentiation and Wnt / β-catenin signaling pathway under tensile stess from the perspective of morphological and molecular biology, enriches the theory of Chinese medicine Bushen Zhuanggu, and objectively evaluates the role of Chinese medicine in promoting osteogenesis.
