课题组采用人舌鳞癌细胞株和通过新鲜外周抗凝血分离并贴壁获取的单核/巨噬细胞,分别在酸性微环境PH值为6.6、6.8和正常条件PH=7.2;缺氧微环境下37℃、O2<1%、常氧微环境下37℃、20% O2、5%CO2进行培养,4小时后,采用MTT法检测单核/巨噬细胞的体外杀瘤活性,ELISA法检测单核巨噬细胞分泌VEGF的情况,同时重复上述细胞培养,提取单核/巨噬细胞总蛋白,采用蛋白组学检测缺氧、酸性和正常微环境下单核巨噬细胞蛋白表达的差异。本研究表明局部缺氧和酸性的微环境下,单核/巨噬细胞对肿瘤细胞的吞噬和杀伤作用减弱,而分泌促进肿瘤生长因子的作用增强,从而更多的参与了肿瘤的生长和转移。本研究还通过蛋白组学的方法来研究是何种功能蛋白蛋白促使单核/巨噬细胞功能改变,目前已经发现数十种差异蛋白,但其意义还需要等待质谱鉴定结果出来后进一步进行分析评价。同时,课题组还进行了与单核/巨噬细胞在口腔癌中浸润机制和功能有关蛋白的免疫组化研究,结果表明在口腔癌中,VCAM-1等参与了单核/巨噬细胞肿瘤中的浸润,其可以通过分泌白介素-8、基质金属蛋白酶-9、环氧氧合酶-2等参与肿瘤的生长和转移。

OBJECTIVE:To investigate the cytotoxicity of monocytes/macrophages against Tca8113 cell and the secretion of vascular endothelial growth factor (VEGF) in hypoxic(O2<1%) and acid(PH=6.6,6.8)microenvironment in vitro. METHODS: Peripheral blood mononuclear cells extracted from healthy person’s blood and cultivated to transform into monocytes/macrophages. Then monocytes/macrophages were cultured with Tca8113 in hypoxic microenvironment (o2<1%,ph=7.2),acid microenvironment(o20%,ph=6.6 and ph=6.8) and in normal microenvironment (20% O2,ph=7.2). The cytotoxicity of monocytes/macrophages against Tca8113 cell was examined by MTT assay . The expression of VEGF was detected by enzyme-link immunoassay (ELISA). RESULTS:The cytotoxicity of monocytes/macrophages against Tca8113 cell in hypoxic and acid microenvironment is lower than in normal environment. The expression of VEGF excreted by mononuclear/macrophage was significantly higher in hypoxic and acid microenvironment than in normal microenvironment. Tens different expression protein spots were screened .CONCLUSION: In hypoxic and acid microenvironment, the cytotoxicity of monocytes/macrophages against Tca8113 cell weakened, the secretion of VEGF was gradually increased. The significance of differential expression protein need be analyzed after mass spectrometry.
