真菌性角膜炎FK是主要的致盲性眼病。课题采用模拟人类自然感染过程的动物FK模型,对FK的发病机制及其提示的防治方案进行了探讨,主要发现包括:巨噬细胞在FK病理改变中发挥保护作用,用GM-CSF促进吞噬细胞对真菌的吞噬作用,有利于病原菌的清除与溃疡愈合;TLR4在FK角膜中与巨噬细胞数量相关;磷脂酶B在FK损害中,仅在真菌黏附角膜的起始阶段促进菌丝穿透细胞膜,而在后期并无明显的毒力作用;整合素家族多种蛋白参与烟曲霉菌与角膜上皮细胞的粘附;采用特异性中和抗体对主要炎性趋化因子和细胞因子进行阻断,有效地减轻FK的病变损害;FK可诱导针对特异性真菌的特异性免疫反应和免疫记忆,在FK中发挥重要的免疫防护作用;应用免疫调节剂CpG寡核苷酸对小鼠角膜真菌感染具有明显的免疫防治作用;动物的免疫状态对小鼠FK病程有重要影响。此外还建立了FK液相核酸芯片新型诊断方法。课题共发表论文27篇,其中SCI收录16篇,总IF指数47.9,并成功申请了国家专利3项,培养了博士、硕士及初级研究人员近20人,并开展了广泛的国际交流。“真菌性角膜炎的应用基础与临床学研究”获山东省科技进步一等奖。

The pathogenesis of fungal keratitis, the leading cause of corneal blindness was investigated and potential therapies were explored using various animal models mimicking nature progress of human FK. Main findings include: macrophages infiltration, TLR4 expression on macrophages, and inflammatory cytokines production by macrophages are involved in FK development, enhancing macrophage function, as well as depleting macrophage or secreted inflammatory cytokines increases resistance of animal to FK; FK induces adaptive immune response and specific immunological memory in mice. Immunostimulatory CpG oligonucleotides manifest protective effects in FK mice. The status of animal significantly affects the progress of FK in them. A novel diagnostic method based on liquid array strategy is developed with both sensitivity and speed of fungal diagnosis greatly increased. Based on the results of this project, totally 27 papers were published including 16 in SCI journals; three national patents were granted, and about 20 doctoral/master degree candidates as well as junior researcher were trained.
