本项目针对雷公藤甲素毒副作用大、成药性差和生物利用度低等问题,以及目前癌症光热治疗中由于肿瘤内部热量分布不均导致的肿瘤治疗失败的现状,我们拟以透明质酸和维生素E琥珀酸酯作为形成胶束的材料,制备一种具有肿瘤CD44受体靶向和氧化还原敏感的多功能聚合物胶束,用于封装水难溶性药物雷公藤甲素和具有强近红外光热转化性能的金纳米棒,并在B超的介导下,实现由近红外光和肿瘤细胞内高浓度谷胱甘肽共同触发的同步肿瘤靶向光热治疗与化学治疗的联合治疗。通过近红外激光的辐照治疗,肿瘤细胞受到了不可逆的光热治疗作用;同时,光热效应和肿瘤细胞内高浓度谷胱甘肽共同触发胶束内雷公藤甲素的释放,产生更强的化学毒性作用;光热治疗和化疗协同作用,双重杀灭肿瘤细胞,实现安全有效的肿瘤靶向治疗,为癌症的个性化治疗开拓了新的思路,有望加快传统中药在纳米医药领域的应用并提高癌症的治愈效果。

The project aimed at the great toxic side effect, the poor druggability and the low bioavailability of triptolide, and the current situation of the failure of tumor therapy caused by uneven tumor heat distribution in the photothermal therapy (PTT) process, we intended to develop a novel multi-functional polymeric micelle with CD44 receptor targeting and redox response using hyaluronic acid (HA) and Vitamin E succinate for encapsulating hydrophobic triptolide and gold nanorods (with strong photothermal conducting property in near-infrared region), to realize the combination therapy of the synchronizing tumor-targeted photothermal therapy and chemotherapy triggered by near-infrared light and high concentration of glutathione in tumor cells under guidance of B ultrasound. Under near-infrared light irradiation, tumor cells have suffered irreversible photothermal injury; Simultaneously, photothermal effect and high concentration of glutathione in tumor cells together triggered the release of triptolide of polymeric micelles, caused stronger chemical toxicity effect; The efficient combination of photothermal therapy and chemotherapy caused a double injury on the tumor cells, further realized safe and effective tumor targeting therapy, which have opened up new ways for personalized treatment of cancer, and contributed to accelerating the application of Traditional Chinese Medicine in nanomedicine field and improving the cure rate of cancer.
