本研究拟以异质性生境中的芦苇为研究对象,选取淡水湿地、咸水湿地、盐渍化土壤等异质性生境设置样地,结合野外群落调查、植物功能性状测量、生境生态因子分析,同时开展典型生境芦苇种群的同质种植园实验及模拟控制条件下的交互移植实验,筛选主导环境因素,确定功能性状对异质性生境的适应性反应,揭示芦苇生态分化的生境驱动及表型适应;采用流式细胞仪和染色体计数进行染色体倍性的鉴定,采用trnT-trnL、rbcL-psaI叶绿体基因序列测序法分析遗传结构,结合移栽前后的DNA甲基化表观遗传变异和表型可塑性测定,检验这些表型变异是源于遗传分化还是环境饰变,检验不同芦苇种群的DNA甲基化重塑能否稳定遗传,厘定遗传因素和环境因素在驱动芦苇生态分化中的相对贡献,揭示异质性生境中芦苇的生态分化及适应性进化机制。研究可为探索生态适应性的遗传进化机制提供重要基础,同时为生态重建、植被恢复以及植物资源保护评价提供科学依据。

Phragmites australis under heterogeneous habitats is taken as the main experimental material in this research. Our research will select different areas, such as fresh water wetlands, salt water wetlands, land with salinity soil, etc. as sample areas to do field survey, plant functional traits measuring and habitat ecological factors analysis; as well as conduct Common garden experiment and analog controlled transplant experiment on P. australis population under typical habitat. In this study, we screen the main environmental factors that affect P. australis and test the adapting response of plant functional traits to heterogenetic habitat, to reveal the relationship between plant functional traits and the growing environment as well as habitat driver of ecological differentiation and phenotypic adaptation. Using Flow Cytometer and chromosome counts, chromosome ploidy can be decided in P. australis samples; based on trnT-trnL and rbcL-psaI chloroplast gene sequencing method, epigenetic variation and phenotypic plasticity before and after planting, we can test whether the phenotypic variation is caused by genotypic differentiation or environmental modification; testing whether gene methylation can stably inherited to evaluate the effect of hereditary factor and environmental factor. And above all, this can reveal the ecological variation and adapted evolution mechanism of P. australis under heterogeneous habitats. This study can provide basic knowledge of genetic evolution mechanism on ecological adaptability, as well as scientific evidence to make ecological reconstruction, revegetation, plant resource protection and evaluation.
