构树是一种具有重要经济利用价值的资源植物。叶片的大小和形状直接影响青贮饲料产量。构树不同发育阶段的叶片形状各异,为典型的异形叶性,而目前构树叶片形态变化机理尚无研究。因此,本项目以野生型和突变体构树为研究材料,以WOX基因为切入点,采用CRISPR/Cas9基因编辑、过表达、转录组测序和生物信息学分析、原位杂交、双分子荧光互补实验等技术,对WOX基因超表达和功能缺失的构树进行系统研究;对不同形状的构树叶片进行结构观察,检测叶片发育过程中的生理生化指标、基因表达数据等,阐明构树异形叶性的形成规律,解析WOX基因在构树叶片形态建成过程中的作用机制。本研究为揭示WOX基因在构树异形叶形态建成中的作用机制积累理论知识,具有重要的科学意义;同时,本研究为提高构树粗蛋白饲料产量,培育饲用杂交构树新品种提供技术指导,具有广阔的应用前景。
英文摘要
The paper mulberry is a kind of important plant resource for utilization. The size and shape of the leaves directly affect Silage Yield. There are many difference about the leaf shape in different development stage of the paper mulberry. However, the leaf morphogenesis and the formation of heterophylly have never been investigated in the paper mulberry. The WOX genes are known to play important roles in regulating the development and morphogenesis of plant leaf. Thus, the key WOX genes played key roles in the leaf development will be screened from the paper mulberry. In order to discern the function of the WOX genes in the leaf morphogenesis and proce, bioinformatics and bimolecular fluorescence complementatio will be adopted in this study. To clarify the developing rule of the paper mulberry’s leaf heterophylly, three kinds of the paper mulberry strains (overexpressed, mutant and control) were used to observe structures of various types leaf, detect the metabolism, physiological processes and gene expression of leaf development, etc. This study will be helpful for the knowledge accumulation of exploring the function of WOX genes involved in the formation of heterophylly. Additionally, it will provides theoretical basis for breeding new cultivar and benefit for the increasing the crude protein feedstuff production of paper mulberry.
