心脑血管疾病，如冠状动脉阻塞等，是影响人类健康的重大疾病。在临床上，药物洗脱支架通过携带抑制平滑肌细胞增殖的药物，广泛用于病人血管成形术后再狭窄的治疗。药物洗脱支架常用的涂层材料通常为易改变其表面特性的功能性聚合物。另外，具有雌激素活性的药物，也具有直接促进血管舒张，并增强内皮愈合等活性。文献表明，雌激素主要是通过结合细胞膜表面的雌激素受体，从而产生的非基因组效应来实现该保护作用。然而，当雌激素结合细胞核内的雌激素核受体从而产生基因组效应时，有会诱导癌症的风险。因此在本方案中，我们以炔雌醇为前体，精心设计了一种可以让雌激素特异性产生非基因组效应的聚合物涂层递送系统，从而安全地发挥其在心血管系统中保护作用，经过一系列物理化学表征测试和细胞研究，我们证明了该新型材料可以刺激内皮细胞心血管保护的明星分子一氧化氮的产生。以上前期实验表明，该雌激素药物支架涂层材料将会在心血管再生医学中发挥重大作用。

Cardiovascular diseases, such as coronary artery occlusion, is a major disease that affecting human health globally. Clinically, drug eluting stents are widely used for the treatment of restenosis after angioplasty in patients with incorporating drugs that inhibit the proliferation of smooth muscle cells. Commonly used coating materials for drug-eluting stents are generally functional polymers that their surface properties are readily modified. In addition, pharmaceutical agents having estrogenic activities were also considered as directly promoting vasodilatation and enhancing endothelial healing. Many studies suggest that these beneficial effects of estrogen on the cardiovascular system are primarily due to the binding of estrogen receptors on the cell membrane, resulting in non-genomic effects. However, when estrogen binds to nuclear estrogen receptors and hereby induces genomic effects, there is a high risk of triggering cancer. Therefore, in this study, we used ethinylestradiol to conjugate to polymer as to develop a novel coating materials, which can specifically estrogen non-genomic effect, in order to exert its protective effect in the cardiovascular system safely, After a series of physical chemistry characterizations and cell studies, we demonstrated that this coating material stimulates the production of nitric oxide, an important molecule in the protection of cardiovascular system. Above preliminary experiments showed that the estrogen based stent coating materials may play a major role in the cardiovascular regeneration medicine.
