锥囊藻科隶属金藻门,是一个广泛分布于淡水水体,对贫营养型水体具有重要指示作用的类群。由于没有细胞壁包围,原生质极易被降解破坏,种类鉴定较为困难,分类系统复杂多变,目前主要依据囊壳形态分类,但前期研究已发现囊壳的有无和形状变化并不是一个稳定的特征。因此本项目将在进行广泛的野外调查和标本采集的基础上,采用形态学、活体培养和分子生物学技术相结合的手段,对我国淡水锥囊藻科植物进行深入系统的、多特征的分类研究,包括(1)利用形态学方法研究锥囊藻的特征,评价关键特征的分类学价值;(2)利用基因序列分析进行分子系统学研究,构建系统发育树,明确该科内各谱系的亲缘关系;(3)综合形态特征和分子数据,澄清科内的系统学类群,构建分类依据和标准,对现有分类系统进行修订,建立锥囊藻科客观有效的分类方法。该项目的研究不仅对全面了解中国锥囊藻科的区系分布现状有重要科学意义,而且对其生物多样性的保护也具有重要的指导作用。
英文摘要
The Dinobryaceae, a family in Chrysophyta, which is wildly distributes in freshwater, is known as an important oligotrophic ecological indicator of relative phosphorus poor and low nutrient water body. However, it is very difficult for identifying its members because the protoplasm of most species is easily degraded as lack of cell wall. The present taxonomic criterion for Dinobryaceae is the shape of the lorica, which is a transparent structure enveloping cells. BUT, this is not reliable as the existence and shape of the lorica is not a stable character indicated in our previous study. To solve this problem, in this study, the systematic taxonomy study will be carried out based on countrywide field investigation and large sampling of Dinobryaceae in China. The reasonable criteria for Dinobryaceae identification will be established by comprehensive analysis of both morphological and molecular data. To achieve this, 1) all potential morphological characters will be evaluated in taxa; 2) phylogenetic trees will be reconstructed based on genetic information from sequence distance of both chloroplast and nucleic conserved genes and inter spaces sequence; 3) the present taxonomic criteria will be revised based on large mount data collected above, and the reasonable taxonomic system will built. Our results will provide comprehensive knowledge of the flora and corresponding distribution of Dinobryaceae in China, as well as the essential scientific support for biodiversity protection.
