脂氧合酶代谢产物茉莉酸(JA)和绿叶性气味(GLV)是重要的抗虫防御信号分子。OsRCI是新发现的虫害诱导水稻脂氧合酶基因,其编码蛋白具C13催化活性,并定位于叶绿体中。我们发现OsRCI基因在抗褐飞虱水稻品系(含Bph1基因)和敏感品系中存在遗传多态性(L--R型和HRRP型)。本项目将构建OsRCI基因的各种突变体,从生物化学、分子生物学、遗传学和反向遗传学角度对其功能进行研究,解析水稻不同型OsRCI基因对水稻虫害诱导抗虫防御反应的影响;明确OsRCI基因在水稻茉莉酸(JA)和绿叶性气味(GLV)合成中的作用,初步解析OsRCI基因调控水稻抗虫防御反应的作用机制;直接将OsRCI作为分子标记基因分析水稻遗传群体,明晰该基因与水稻Bph1基因的关系。本研究有望增加对水稻抗虫防御反应分子机理的认识,研究结果可为抗虫水稻品种的选育提供分子理论依据,或者可作为抗虫基因直接应用于农业生产。

The Jasmonic acid (JA) and Green leaves volatiles (GLV), the products of lipoxygenase pathway, are the most important signal moleculars of plant defense signaling transduction. OsRCI is one new herbivore-induced rice lipoxygenase gene, the protein of OsRCI is located in Chloroplast and appears C13-LOX catalysis activity. We found that OsRCI gene showed two genetic polymorphisms (L--R type and HRRP type) in rice brown planthopper non- and resistant rice varieties. Various OsRCI mutants will be constructed, their functions will be studied from different fields as biochemistry, molecular biology, genetics and reverse genetics in order to revealing 1) the roles of two types of rice OsRCI gene (L--R type and HRRP type) in synthesis of Jasmonic acid (JA) and Green leaves volatiles (GLV); 2) the mechanism of OsRCI in regulating rice herbivore induced defense responses; 3) discovering the relationship between OsRCI gene and rice Bph1 gene. This study may deepen the understanding the function of rice lipoxygenase family in plant resistance, and the molecular mechanism of rice induced defense response. The findings are expected to provide a theoretical basis for the molecular breeding of insect-resistant rice variety, or those genes may be directly used as insect-resistant genes in application of agricultural production.
