已知癫痫反复发作可导致神经元损害,出现脑功能受损症状,影响认知功能。前期临床和实验研究已证实,以“益肾填精法”为主的茸菖胶囊对认知障碍的癫痫患儿或致痫大鼠均有改善作用,在保护受损神经细胞方面具有明显优势。故提出如下假说:“以益肾填精法为主的茸菖胶囊预处理可有效改善致痫大鼠海马微环境,包括BBB功能及相关炎性因子的表达,达到防治癫痫后神经损伤和认知损害的作用”。以海马组织损伤为切入点,以炎性反应及BBB功能改变为主线,以SD大鼠建立戊四唑点燃模型,分正常对照组、模型组、茸菖胶囊预处理组和西药对照组。造模同时药物干预8周后检测:①大鼠反复惊厥行为学及海马组织改变(DNA片段分析);②对BBB功能的影响研究:超微结构观察、通透性、claudin-5的表达;③对炎症反应的影响:IL-1β、HMGB1以及TGFβ、CCL4表达。阐明益肾填精法预处理的防治癫痫后神经损伤机制,为其临床应用提供实验依据。

Repeated attacks of epilepsy in children can lead to neuronal damage, impacts.cognitive function. Earlier researches had confirmed that enriching kidney essence method based on in Rong Chang capsule capsule can improve cognitive impairment for children with epilepsy or induced epileptic rats ,and had obvious advantages in protecting the damaged nerve cells.Therefore, put forward the following hypothesis: “The enriching kidney essence method based Rong Chang capsule pre-processed can effectively improve the micro environment of hippocampus of epileptic rats, including BBB function and related inflammatory factor expression, achieved the prevention and treatment of epilepsy after nerve injury and cognitive impairment effect.”With hippocampal tissue damage as the breakthrough point, inflammatory reaction and BBB function change as the main line, pentylenetetrazol (PTZ)-kindled model used SD rats, which divided into Divided into normal control group, model group, Rong Chang capsule pretreatment group and Western medicine group.Intervention in 8 weeks after the test: (1) repeated seizures in rats behavior and organizational change hippocampus (DNA fragment analysis);(2) research on the influence of BBB function: ultrastructure observation, permeability test, the expression of claudin-5;(3) the impact on the inflammatory response: the expression of IL-1β, HMGB1 and TGFβ, CCL4 .To clarify mechanism of prevention and treatment of epilepsy after nerve injury by the enriching kidney essence method pretreatment, providing experimental basis for its clinical application.
