中药复方所产生的生物效应，是其所含的化学成分通过多途径、多靶标的协同作用而产生的结果，揭示中药复方作用机制一直是中药现代化研究中的关键问题。本项目以益气活血方（脑心通方）抗脑缺血作用的分子机制研究为范例，采用多层异质网络分析与深度高通量蛋白质组技术相结合的筛选和辨识方法，在挖掘中药复方多成分多靶标有效组合的基础上，按照“网络构建—关联计算—高通量蛋白质组筛选—数学建模—实验验证—分子机制”等六个步骤，将计算筛选与实证研究有机结合，揭示益气活血方（脑心通方）抗脑缺血的作用机制，为中药复方多成分多靶标的有效协同组合的作用机制研究提供范式。.本项目将多层异质网络分析与深度高通量蛋白质组技术有机结合，既提高了中药复方作用机制的实证研究的精确性，也突破了网络构建与计算筛选后的实验验证面临的难点，为中药复方作用的分子机制研究提供新思路、新技术和新方法。

The biological effects of Chinese formula result from synergistic interactions of multiple compounds contained in Chinese medicine due to its various pathways and targets. It is really a critical problem to reveal mechanisms of Chinese formula to facilitate modernization of traditional Chinese medicine (TCM). This project takes the study on mechanisms of action of Chinese formula Yi-Qi-Huo-Xue(BNC formula) in the treatment of cerebral ischemia as an example, using heterogeneous network analysis combined with high-throughput proteomics technology to provide a paradigm on the mechanism of synergistic interactions exerted by multiple compounds and targets contained in Chinese medicine. This method is based on the data mining of combination between multiple compounds and targets, in terms of “network construction- contextual computing- high-throughput protein screening- mathematical modeling- experimental validation- molecular mechanism”, achieving organic bond of computing screening and empirical study, uncovering the mechanisms of action of Yi-Qi-Huo-Xue(BNC formula) in the treatment of cerebral ischemia..This project combined heterogeneous network analysis and high-throughput proteomics technology organically, not only improved the accuracy of action mechanism in Chinese medicine of empirical study, but also made a breakthrough to address problems from experimental validation behind network construction and contextual computing, provide new thought, new technique and new strategy for study on molecular mechanism of Chinese formula.
