多囊卵巢综合征(PCOS)是以雄激素过多和持续无排卵为主的生殖障碍性疾病,其雄激素成因与发病机制复杂,临床缺乏特效中药持久阻断高雄激素以及反复波动造成的PCOS妊娠后胚胎丢失的病理状态。卵巢局部调控因子FKBP52是FKBP家族中参与生殖调控的最重要一员,其能够通过与伴侣蛋白结合促进糖皮质激素、雄激素等多种类固醇激素-受体复合物的形成,调控目的基因的表达。基因芯片技术发现PCOS患者FKBP52表达异常,影响胚胎植入,造成反复流产,但具体形成机制并不清楚。本项目试图以名医中药验方菟丝子的提取物金丝桃苷干预FKBP52表达而产生对雄激素-雄激素受体结合及其后MAPK/Erk信号传导通路的影响,探讨金丝桃苷对PCOS雄激素成因与发病机制的作用,阐明补肾中药改善雄激素代谢、重建PCOS月经周期、促进妊娠的潜在机制。

Polycystic ovary syndrome (PCOS), as one of the reproductive disorders, is mainly charactered by hyperandrogenism and chronic anovulation with complicated aetiology and pathogenesis. However, there is still no specific remedy to reduce hyperandrogen stably and reverse the pathological state of fetal loss in PCOS. FKBP52 is the most important member of FKBP family to involve in the regulation of reproduction as an ovarian local regulatory cytokine, and it regulates the expression of genes by controlled the formation of glucocorticoid and androgen combined with steroid hormone receptors. Detection of abnormal FKBP52 in PCOS is associated with embryo implantation failure and recurrent abortion by gene chips technology, but the specific regulatory mechanism is not clear. This project attempts to explore the effect of hyperoside derived from semen cuscutae on the expressions of androgen, androgen receptor and MAPK/Erk signaling pathway throng FKBP52, in order to clarify the potential mechanism of Chinese herbs for invigorating the kidney on improving the metabolism of androgen, reconstructing menstrual cycle and promoting pregnancy.
