沙苑子为传统补阳药,可通过温肾阳,助气化达到化浊降脂之功。前期研究表明,沙苑子对去势高脂大鼠具有显著的降TG作用,对雌激素虽无明显升高,但却能显著增加肝脏ER-α和ER-β mRAN的表达。因此,本课题以ER为切入点,以雌激素为对照,观察沙苑子总黄酮对去势高脂大鼠肝脏TG合成、转运、水解的影响,通过检测调控TG合成SREBP-1c-FAS-ACC-GPAT和PPARα-CPT-1α-ACO,转运HNF4α-MTP-ApoB100-VLDL和水解PPARγ-ATGL-HSL等通路,分析沙苑子可能的降脂机制;同时采用体外培养肝细胞,利用ER阻断剂,观察沙苑子对以上通路的影响及其与ER的相关性,比较沙苑子与雌激素对肝脏TG代谢作用途径和靶点的异同,分析中药补阳药与雌激素替代疗法的不同之处,探讨沙苑子温阳化浊的调脂机制,揭示补阳药药性理论的科学内涵,并阐明中药补阳药调节血脂的特色和优势。

As the conventional Yang-Reinforcing herbs, SHAYUANZI is able to resolving turbidity and regulating blood lipid for warming kidney yang and assisting qi transformation. Previous research has shown that SHAYUANZI could decrease the level of TG of the ovariectomized hyperlipidemia rats, at the same time the mRNA expression of liver ER-α and ER-β are significantly decreased. So this study will take the ER as the breakthrough point and take estrogen as the control, and observe the effect of total flavonoids of SHAYUANZI on liver TG synthesis, transport and hydrolysis in ovariectomized hyperlipidemia rats. To analyze the possible mechanism of SHAYUANZI in regulating blood lipid by detecting channels as follow: TG synthetic SREBP-1c-FAS-ACC-GPAT and PPARα-CPT-1α-ACO, TG transporting HNF4α-MTP-ApoB100-VLDL and TG hydrolysis PPARγ-ATGL-HSL. At the same time we use hepatocytes cultured in vitro, using ER blocking agent, to observe the effect of SHAYUANZI on the above channels and its correlation with ER, comparing the differences between estrogen and SHAYUANZI, to investigate the mechanism of warming yang and regulating blood lipid of SHAYUANZI, to reveal scientific connotation of pharmacological of reinforce yang herbs and to clarify the characteristics and advantages of reinforce yang herbs in regulating blood lipid.
