皮肤皱纹是皮肤衰老的临床表现之一,影响美观,是医学美容关注的热点。研究发现氧化应激伴增龄而增强,可增加基质金属蛋白酶表达,降解皮肤胶原纤维,导致皮肤皱纹形成。中医认为肾精亏虚是皮肤衰老、皱纹形成的重要原因,补肾益精是延缓衰老、减轻皱纹的有效治法。院内制剂养颜青娥丸是国家名老中医经验方,临床应用多年,具有补肾益精之功效,是体现该治法的方药,并能抑制氧化应激,改善皮肤皱纹,但作用机制有待深入。本项目基于已结题青年基金成果(β-catenin抑制p16INK4a,减轻氧化应激,延缓皮肤衰老),建立小鼠皮肤光老化模型,同时养颜青娥丸干预,并培养模型鼠原代皮肤成纤维细胞,借助基因沉默、基因敲除等手段,通过免疫荧光、分子生物学、透射电镜等技术检测基于β-catenin抑制p16INK4a介导氧化应激,调控衰老等相关指标,旨在研究养颜青娥丸对皮肤光老化皱纹的作用机制,为养颜青娥丸的临床应用提供科学依据。

An important indicator about severity of skin aging is wrinkle. Research found out that oxidative stress injury which enhanced with aging increased the express of active oxygen and matrix metalloproteinase, with degraded collagen in dermis, as a result, causing skin aging and wrinkle formation. Due to the theory of Chinese medicine, deficiency of kidney and essence is an important incentive for the formation of wrinkle, as well as skin aging. While in clinical practice, kidney-nourishing and essence strengthening is one effective treatment of delaying skin aging and reducing the wrinkles. A Chinese medicine named Yang Yan Qing E Wan is a classic Chinese prescription for such application. With the function of nourishing kidney and strengthen essence, it can effectively treat the skin wrinkles, how ever, pharmacology and mechanism of this medicine had yet to be further developed. Based on the pervious research by us, we would build three mouse photo aging models: Ultraviolet, Ultraviolet with β-catenin silencing, Ultraviolet with β-catenin knockdown. At the same time, we pretreat the models by Yang Yan Qing E Wan. To acquire cultured rat skin fibroblasts. We detect the mark of apoptosis and aging during the β-catenin mediated skin aging in primary fibroblast which getting from the mouse models. These researches should explore the target and molecular mechanism of Yang Yan Qing E Wan in reducing the skin aging wrinkle formation.
