基于中药复方愈口宁对复发性口疮(RAU)心脾积热证的临床疗效与影响机体免疫的作用,本课题拟围绕调节性B细胞(Bregs)功能相关的PI3K/Akt信号通路,探讨愈口宁治疗RAU(心脾积热证)的药理机制及其清热解毒治法特点。首先观察愈口宁及其清热解毒拆方对模型大鼠口腔溃疡、T淋巴细胞和IgA、IgG、IgM的影响。继之围绕Bregs活化中PI3K/Akt信号通路,观察药物对EGF、EGFR、Bregs、细胞因子等影响。最后分析其清热解毒治法与免疫调节的相关性。从而验证“愈口宁通过调控EGF和EGFR,激活PI3K/Akt信号通路,活化Bregs,调节细胞免疫和体液免疫,促进溃疡愈合并减少复发;其中清热解毒治法药物对于Bregs活化及其免疫调节发挥关键作用”的科学假说,阐明愈口宁治疗RAU(心脾积热证)的主要机理,促进该药的临床应用与深入研发,提高中医药治疗口腔黏膜疾病的基础研究水平。

Chinese herbal Yukouning showed good clinical effect and immune regulative effect on recurrent aphthous ulcer (RAU) patients with brimming heat of heart and spleen (BHHS type). This study will focus on the mechanism of Yukouning on immune pharmacology and its characteristic of disassemble prescription around regulatory B cells (Bregs) function-associated PI3K/Akt signaling pathway in RAU (BHHS type) rat. Firstly, oral ulcer, T lymphocytes, IgA, IgG, IgM will be tested. Secondly, around the PI3K/Akt signaling pathway in the activation of Bregs, the level of EGF and EGFR, Bregs, IL-10, TGF-β and Tregs will be investigated. At last, the relationship between immune regulation and clearing heat-toxin method will be analyzed. The scientific hypotheses of Yukouning promoting ulcer healing and reducing the risk of recurrence on RAU(BHHS type) by means of activating Bregs, adjusting cellular and humoral immunity after regulating the level of EGF and EGFR through PI3K/Akt signaling pathway as well as the key role of clearing heat-toxin drugs in Yukouning together will be verified. Furthermore, Yukouning pharmacological mechanism will be clarified and be popularized as drug development, and the basic researching levels of TCM treatment on oral mucosal diseases would be improved.
