腐败希瓦氏菌是鱼、贝类以及甲壳类等富含蛋白质水产品的特定腐败菌。抑制希瓦氏菌是解决水产品加工与贮藏过程中腐败问题的关键之一。利用生物保鲜剂抑制希瓦氏菌,可以显著延长鱼、贝类以及甲壳类等富含蛋白质水产品的货架期。因此,寻找有效的抑制希瓦氏菌的生物保鲜剂是解决水产品加工与贮藏过程中腐败问题的重要手段。.贝类凝集素是其天然免疫因子,通常具有抑制微生物的活性,是一种潜在的抑菌剂来源。前期研究发现了菲律宾蛤仔凝集素对希瓦氏菌产生的抑制作用。本项目拟在前期研究基础上,深入研究菲律宾蛤仔凝集素对希瓦氏菌产生抑制作用的机制,系统地解析出菲律宾蛤仔凝集素与希瓦氏菌细胞膜及膜上分子的相互作用,进而利用蛋白质组技术及代谢组学技术揭示出菲律宾蛤仔凝集素干扰希瓦氏菌代谢途径产生抑制作用的机理。研究结果有助于深入认识凝集素抑制细菌的机理,为获得贝类凝集素类抑菌剂提供基础数据。

Shewanella putrefaciens was specific spoilage organism (SSO) of protein-rich fish, shellfish and crustacean. The growth inhibition of S. putrefaciens was a key way for resolving putrid problem in aquatic products. Using biological bacteriostatic agent could significantly prolong aquatic products shelf life of protein-rich fish, shellfish and crustacean. Therefore, the finding of effective biological preservatives against S. putrefaciens was an important means of resolving putrid problem during processing and storage of aquatic products..Lectin of shellfishes is one of innate immunity factors. It is associated with inhibition activity to microorganisms. Therefore lectins of shellfish were potential sources of biological bacteriostatic agent. Previous studies showed that short-neck clam Ruditapes philippinarum lectin (MCL-T) could inhibit the growth of S. putrefaciens. In this work, on the basic of previous results the effects of R. philippinarum lectins on the membrane and membrane molecules of S. putrefaciens will be investigated. And then proteomics and metabolomics are applied to investigate the mechanism changes of S. putrefaciens after inhibition effect of R. philippinarum lectin. It will lay the foundation for understanding of the mechanism of inhibition of S. putrefaciens and developing biological bacteriostatic agent from shellfish lectins.
