丹参茎叶在清代医籍已有药用记载,文献及本课题组前期研究提示其具有改善血液循环和抗炎活性,且含有的多元物质体现出独特的效应特点。据此提出“丹参茎叶酚酸类、黄酮类、三萜类多元效应物质可通过调控炎症反应及相关信号通路实现对血液循环障碍的调节作用”的科学问题,拟开展研究工作:(1)基于色谱—质谱多级联用技术对丹参茎叶多元化学物质进行表征分析;(2)基于血瘀、炎症损伤等模式生物体内外模型评价丹参茎叶的生物效应;(3)基于体内过程揭示其体内效应物质及其代谢转化,并应用适宜数学模型对其谱-效关系进行分析,明确多元效应物质组成与量效关系;(4)基于代谢组学阐明血瘀、炎症损伤所致的代谢变化和代谢通路异常以及丹参茎叶的干预效应;(5)基于血小板活化聚集、血管内皮功能、炎症因子及相关信号通路等从分子水平揭示其各类化学成分/组分改善血液循环、抗炎作用的分子机制。以期为丹参茎叶药用价值发现和资源化利用提供科学依据。

英文摘要

Salvia miltiorrhiza stems and leaves have been recorded with great medicinal value from Qing Dynasty. Modern researches suggest its anti-inflammatory activity and effect on improving blood circulation due to multiple active components with unique characteristics of effect, based on which, a scientific issue that “phenolic acids, flavonoids, triterpenoids in Salvia miltiorrhiza stems and leaves can prevent blood circulation disorders via regulating inflammation and relative signaling pathway” is put forward. Research works in preparation are as follows: the first is to analyze chemical composition in Salvia miltiorrhiza stems and leaves via multistage chromatography-mass spectrometry detection technique. The second is to estimate biological effect of Salvia miltiorrhiza stems and leaves based on not only blood stasis and inflammatory injury animal models but also issues, cellular and molecular levels. The third is to assess the spectrum-effect relationship via correlation analysis of mathematical model, confirming the multiple active constituents and their dose-effect relationship and in further revealing active compounds and their metabolic biotransformation in vivo. The forth is to expose the regulating effect of Salvia miltiorrhiza stems and leaves on metabolic profiling, bio-target and metabolic pathway based on blood stasis and inflammatory injury animal models using metabolomics technology. The fifth is to illuminate the action mechanism of multiple active constituents on improving blood circulation and inflammatory from molecular levels. The ultimate aim is to provide reference for medicinal value development and resource utilization.
