赤拟谷盗Tribolium castaneum是世界性的重要的储粮害虫,由于其强烈的磷化氢抗性而被认为是最难防治的储粮害虫之一。本项目以赤拟谷盗为研究对象,在前期研究的基础上采用基因转录组、RNAi和RT-qPCR技术鉴定与赤拟谷盗信息素生物合成途径中关键基因及其这些基因对信息素生物合成途径中的功能,研究内分泌激素保幼激素和蜕皮激素及其作用相关的基因如受体基因Met和EcR、USP及转录因子Kr-h1、Br等基因沉默后对雄虫释放信息素的影响,研究保幼激素和蜕皮激素及其作用相关基因沉默后对信息素生物合成关键酶基因表达的影响。从化学分析到生理测定然后深入到基因调控,首次将赤拟谷盗信息素生物合成与激素调控研究结合起来,确定赤拟谷盗信息素生物的关键基因及激素调控机制。不仅可以补充昆虫生理生化理论,对综合利用昆虫信息素和激素开发新的杀虫剂具有重要的实践参考意义。

Tribolium castaneum (Coleoptera: Tanebrionidae) is a major cosmopolitan pest of stored agricultural products worldwide. It was considered as one of the most difficult to control stored product insects owing to high resistance against phosphine. Based on the early research, the following research will be carried out: The key genes and their function on pheromone biosynthesis pathway of Tribolium castaneum will be identified by transcriptome, RNA interference and realtime-PCR technologies, The effect exogenous endrocine such as junvile hormone and ecdysone on the pheromone production and the expression of key gens on pheromone biosynthesis pathway. The effect of knockdown genes the junvile hormone receptor and action such as Met, EcR/USP, Kr-h1 on the pheromone production and the expression of key gens on pheromone biosynthesis pathway. From chemistry analysis to physiological test and more to genes regulation, this is first time combination of research on endrocine regulation and pheromone biosynthesis is carried out in this project. The expected results would be elucidate the science problem about the key genes on key gens on pheromone biosynthesis pathway of Tribolium castaneum and their regulation mechanism by endrocine. The projects not only will be a complementary for insect physiology and biochemistry, but also have significance practical reference meaning.
