痰湿肥胖被认为是影响高血压发展及预后的重要影响因素。因为动物模型的缺乏以及临床试验实施的难度,相关机制研究进展甚微。由于高血压是一种慢性疾病,长时间和连续性的研究对于探讨其影响因素转化演变的机制至关重要,发现痰湿肥胖转化演变的分子机制,对于控制影响因素与治疗高血压具有重要临床意义。前期研究中,本项目组自2010年集中对一个近4000人的人群进行了连续跟踪和样品采集,现在保存有200余例伴有或不伴有痰湿肥胖转化演变的高血压患者连续3年以上的完整医学信息和血尿样本。本研究拟在此基础上采用代谢组学方法对这些患者的血尿样本进行分析,并将相关结果与患者的临床信息整合进行关联分析,找出痰湿肥胖因素影响高血压发展的可能作用靶点。进而应用动态网络生物标志物分析技术,发现相关节点信号和靶标,从而阐明痰湿肥胖的转化演变影响高血压发展的分子机制,为后续探索祛痰除湿基础上的个体化治疗高血压的网络靶点提供科学基础。

The role of obesity with Chinese medicine pattern of phlegm-dampness (PD) in the pathogenesis of hypertension (HTN) is considered as an important factor. However, many animal models for obesity with PD represent clinical manifestations just inaccurately. And some clinical trials are methodologically flawed which makes it difficult to evaluate long term change in obesity with PD on HTN. Therefore, little is known about the molecular mechanism of change in obesity with PD on HTN. To examine changes of risk factors for developing HTN, long term follow up data is becoming increasingly important. The molecular mechanisms of obesity with PD and its changes on HTN have important clinical values in the management of HTN. A 2010-2015 four thousands samples community-based cohort has been established. More than 200 blood and urine samples have been taken before and after for three years period. Among them, participants are categorized into HTN patients with or without change in obesity with PD. With these samples, our aim was to use metabolomics to investigate the metabolic impact of changes in obesity with PD on HTN. Use dynamic network biomarkers analyses with clinical informatics to identify potential biomarkers for exhibit PH individualize treatment for patients with HTN.
