本课题拟选择目前广泛应用的几种纳米材料,以游仆类纤毛虫为主要实验对象,采用常规急性和慢性毒理实验以及MTT细胞活性测定方法,确定每种纳米材料在不同粒径和浓度条件下,对纤毛虫细胞的毒性效应;以扫描电镜和透射电镜为主要手段,并结合酶细胞化学和能谱分析,对多种纳米材料对纤毛虫毒性效应的细胞学机制进行深入研究,以期丰富原生动物毒理学和细胞学研究资料,完善纳米材料毒性效应评价指标体系,为高等动物纳米材料毒理研究提供参考和依据。
英文摘要
This project intend to choose several kinds of nano materials used widely, take Euplotes ciliates as the object, using conventional methods of the acute and chronic toxicity test and MTT cell activity test, in order to determine the toxic effect of different particle size and concentration nano materials on ciliates cells.To study in-depth on the cytological mechanism of nano materials toxic effects by using scanning electron microscopy and transmission electron microscopy as the main means combined with the enzyme cytochemistry and spectrum analysis. So that enrich the protozoan toxicology and cytology research data, improve the evaluation index system of nanomaterials toxicity, and provide reference for the study of higher animal nano materials toxicology.
