花粉外壁的形成是花粉发育的重要组成部分,对植物繁殖至关重要,作为该过程的核心调控因子,MYB80已被用于构建雄性不育系应用于作物杂交育种,但其上游调控通路仍不清楚。本项目以位于BrMYB80基因区域并与之一样在白菜四分体时期花药特异表达的三个lncRNA为研究对象,通过观察它们表达抑制和过表达转基因植株表型并与BrMYB80相应转基因植株表型作对比,分析它们对下游基因及BrMYB80表达的影响,进而利用RNA纯化的染色质分离、RNA酶保护实验等方法鉴定它们是与DNA、RNA还是蛋白质结合起作用,以此获知它们在花粉外壁发育中的生物学功能、作用机制以及在BrMYB80表达调控中的作用。为梳理MYB80调控通路及花粉外壁发育的分子机理,也为解析lncRNA在花粉发育中的作用,最终充分认识花粉发育基因表达调控网络,推进对作物雄性不育机制的理解及重要功能基因的利用提供理论基础。
英文摘要
The formation of the outer pollen exine is essential for the protection of the male gametophyte and plant reproduction. MYB80, a R2R3-type MYB transcription factor, is required for exine formation, and systems involving manipulation of MYB80 function have been developed for the production of F1 hybrid seeds. However, the regulation of the expression of MYB80 is unknown yet. Taking the advantage of transcriptome sequence, three lncRNAs co-expressed with BrMYB80 are found at the regions of BrMYB80 in Brassica rapa. In this program, we try to explain the biology functions, molecular mechanisms and the relationships to BrMYB80 of these three lncRNAs through observing the phenotypes of the expression defected and over-expression lncRNA plants, defining the down-stream genes and the regulation for them on the expression of mRNA and proteins of BrMYB80 by RNA-seq and Western-blot, detecting the interactions between the lncRNAs and DNA, RNA sequences, so do proteins through ChIRP, RNA pull-down and RNase protection assays. These researches will do a great favor on understanding the MYB80 regulation pathway in exine formation, get us to know more about the function of lncRNA played in pollen development and as well as the mechanism underline the male sterility.
