小麦是我国第三大粮食作物,土壤盐渍化是影响小麦产量的重要非生物逆境因素。我们前期通过小麦盐胁迫前后基因差异表达谱分析,鉴定出一个盐胁迫诱导上调表达的组蛋白乙酰转移酶相关基因TaGCN5。本申请课题将以TaGCN5基因为研究对象,从基因表达、转基因功能鉴定及下游候选靶基因的筛选等不同层面上开展研究,为解析小麦耐盐性的表观遗传调控机理奠定基础。具体研究目标为:分析TaGCN5部分同源基因在不同倍性小麦盐胁迫前后的表达变化,获得小麦中该基因的超表达和RNAi转基因株系,查明转基因植株与受体基因型在表型以及耐盐性指标上的差异,筛选出2-3个重要的下游候选基因,初步解析TaGCN5在小麦盐胁迫过程中参与的调控途径。发表论文2-4篇,培养研究生2名。

Wheat (Triticum aestivum L.) is the main crop in China and its yield and performance are severely affected by high salt concentrations in soils, but the underlying mechanisms for salt tolerance in wheat is still need to be elucidated. Recently, a mount of salt stress related genes were identified in wheat by RNA-seq. Although epigenetic mechanisms are crucial to transcriptional regulation of gene expression, their involvement in response to salt stress is less clear. In this study, the main objectives are to figure out how histone acetyltransferase TaGCN5 functions in responsibility to salt stress in wheat. Our specific aims are: 1) detect the TaGCN5 homoeolog-specific gene expression pattern under salt stress. 2) Figure out the biological function of TaGCN5 with overexpression and suppressed its expression in wheat. 3) Compare the salt tolerance between transgenic line and wild type. 4) Determine the downstream genes of TaGCN5 by ChIP-Seq. In summary, the expected data will provide some evidence that TaGCN5 might act as an important factor to salt stress in wheat.
