The applicant has been actively pursuing frontier basic and clinical neuroimaging studies. We have elucidated the intricate relationship between brain functional connectivity and polysynaptic pathway using magnetic resonance imaging techniques. The main research accomplishments include: (1) We have investigated the intersubject variability in connectivity using resting-state functional MRI (fMRI). This study provides a possible linkage between the diversity of human abilities and evolutionary expansion of specific brain regions. The finding is important for interpreting results from functional connectivity studies because conventional brain imaging studies often average data from multiple subjects, an approach that might mischaracterize regional differences. In addition, as clinical practice moves ever closer to the goal of individualized diagnosis and therapy, knowing the distribution of individual differences in brain connectivity is likely important. (2) We have demonstrated that functional connectivity between the cerebral cortex and the contralateral cerebellum is disrupted after the pons infarct. Our study further investigated functions of the cerebelllar intrinsic connection within the whole brain networks. The results provide direct evidence that functional connectivity depends on intact connections within a specific polysynaptic pathway. The results also raised the possibility of using functional connectivity, as a clinical measure, to study behavioral impairment and predict recovery. (3) On the basis of our prior work, we applied functional MRI technique to address key problems: development of novel techniques for accurate diagnosis of cerebral ischemic penumbra, study of the protective effect and mechanism of normobaric oxygen (NBO) in acute ischemia and mechanisms of acupuncture for treatment of dementia. The applicant has published 35 SCI papers in total, which have been cited 428 times, being the first author or corresponding author for 12 articles, including Neuron, Journal of Neuroscience, and Neuroimage, etc. Among these papers, the highest impact factor is 15.982. The applicant intends to apply fMRI study to monitor reorganization of cerebral networks after cerebral infarction with impaired motor function.
