英文摘要
The applicant’s research efforts mainly focus on liver surgery, liver transplantation and translational research of clinical immune therapy. Major achievements are as followings: 1) Applicant combined fundamental research on regulatory T cells (Tregs) and immune induction therapy after liver transplantation for the first time (F1000 score 8). In order to settle the obstacles that Tregs were scarce in vivo and functionally unstable after expansion in vitro, applicant first reported the method of inducing Tregs in vitro using TGF-βand its signal pathways. Up to now, eight patients have successfully received Treg therapy to induce immune tolerance after liver transplantation. Relevant papers were published on PNAS et al. 2) Applicant first systematically studied the signaling pathways of endoplasmic reticulum stress (ER-stress) in liver ischemia reperfusion injury (IRI) and the clinical intervention strategies. Liver graft circulatory support system was investigated and developed to suppress ER-stress, thus improving the quality of liver graft. Three ER-stress pathways during liver IRI were systematically studied to develop possible intervention strategies. Research results were published on Am J Transplant. During the research, 32 papers were published in SCI-indexed journals, and total non-self citations are 273 . Applicant also achieved the honor of “Highly-Cited Chinese Researcher 2014” in the field of medicine released by Elsevier. Upon these foundations, further studies will be performed to analyse the impact of miRNA-mRNA regulatory network on the function of naturally occurring regulatory T cell (nTreg), and to find potential miRNAs which could be utilized for clinical use.
