细胞自噬在RSV感染哮喘气道炎症损伤中发挥重要作用,PI3K/Akt/mTOR是细胞自噬调控的关键信号通路。前期研究发现清肺口服液可以下调RSV感染呼吸道上皮细胞自噬关键分子LC3-Ⅱ及哮喘相关自噬分子Atg5蛋白的表达,从而下调炎症因子表达,提示清肺口服液可以抑制细胞自噬而减轻气道炎症。鉴此,本项目拟构建体内外RSV感染气道损伤模型,使用电镜、免疫组化、小鼠肺功能仪、RT-PCR、Western blot方法观察清肺口服液对体内模型细胞自噬定位表达、肺功能、气道炎症因子及关键信号分子的影响;使用激光共聚焦、基因克隆、信号通路阻滞、免疫共沉淀技术,研究清肺口服液通过PI3K/Akt/mTOR通路调控呼吸道上皮细胞自噬而减轻气道炎症,揭示清肺口服液调节RSV感染哮喘后气道炎症损伤修复作用的机制。为中医药治疗RSV感染哮喘儿童提供理论依据和新的治疗策略。

Autophagy plays an important role in asthma of children’s airway inflammation injury with Respiratory Syncytial Rirus (RSV) infection,PI3K/Akt/ mTOR signaling pathway is a key signal pathway regulator of cell autophagy. Previous studies has found that Qingfei Oral Liquid can decrease LC3-Ⅱprotein expression of autophagy and Atg5 expression of asthma-related-autophagy molecules in human’s airway epithelial cell with RSV infection, which can reduced expression of inflammatory cytokines, suggesting Qingfei oral liquid can mitigated airway inflammation by inhibiting autophagy. Thus, the project intends to build RSV infection airway modelin vivo and vitro, we plan to observe that Qingfei Oral Liquid can reduce autophagosome of lung in mice by using electron microscopy and immunohistochemistry, lung function was observed by Buxco RC in mice. The effect of Qingfei Oral Liquid on the expression of airway inflammation cytokines and key signaling molecule by RT-PCR and Western blot. On the cellular level, we will research changes of cell macrophage morphology and the expression of PI3K, Akt, mTOR, LC3-Ⅱ, baclin-1 protein, by confocal laser,gene cloning, signaling pathways blocking, co-immunoprecipitation etc. Through analysis the datas, we probes into the mechanism of intervention RSV infection in asthma of children’s airway infection injury with Qingfei Oral Liquid. And provide a theoretical basis for preventing and/or treatment airway inflammation of asthma of children who were infected with RSV in Traditional Chinese Medicine.
