过敏性疾病日渐成为儿科临床面临的重要课题,现代医学尚缺乏理想防治方法。中医健脾化湿法防治过敏性疾病有确切疗效,但其机制尚未明确。最近研究提示,iNKT细胞介导的肠道微生物-黏膜免疫失调,与过敏性疾病炎症反应的发生密切相关。我们猜想,健脾化湿法早期干预可通过调节iNKT细胞介导的肠道微生物-黏膜免疫,减轻过敏性疾病炎症反应的发生。本项目拟分别以过敏性结肠炎和支气管哮喘模型小鼠为研究对象,研究健脾化湿法的代表方剂——参苓白术散,早期干预以减轻过敏性结肠炎和支气管哮喘炎症反应的作用,及其与iNKT细胞的关系。还拟结合宏基因组学、DNA甲基化等实验方法,验证参苓白术散是否通过肠道微生物—CXCL 16—CXCR 6的途径,影响体内iNKT细胞水平,并分析其可能作用的关键微生物及其功能基因。研究结果将为深入的疗效机理研究建立基础,并深化对中医“肺脾相关”理论的认识和“培土生金”治法的应用。

Allergic diseases have become important issues in clinical pediatrics, and modern medicine is still lack of ideal method. Traditional Chinese Medicine has exact effect on allergic disease of children with the method of strengthening spleen and eliminating dampness, while the mechanism is not clear yet. Recent researches suggested that iNKT cells, which mediate the intestinal microbiota and mucosal immunity, are closely related to the inflammation of allergic diseases. We speculate that early intervention with the method of strengthening spleen and eliminating dampness may help reduce inflammation of allergic diseases, through iNKT cells which mediating the intestinal microbiota and mucosal immunity. In this project, with mice models of allergic colitis and bronchial asthma as objects of the study, we will explore the early intervention with Shenlingbaizhu powder, the representative prescription of method of strengthening spleen and eliminating dampness, in relieving the inflammation of allergic colitis and bronchial asthma, and the roll of iNKT cells in it. Coupled with methods of metagenomics and DNA methylation, the pathway of intestinal microbiota- CXCL 16-CXCR 6 will be studied to testify the mechanism of Shenlingbaizhu powder on iNKT cells, and key microbes and functional genes will also be filtered out. The results will establish the foundation for further in-depth study on effect mechanism, and deepen the understanding of the theory of ‘lung related to spleen’and the application of ‘strengthening earth to generate metal’.
