本研究以附加结球甘蓝2号染色体(含有BoFLC3基因)的大白菜单体异附加系后代为材料,通过4代回交和游离小孢子培养,结合甘蓝相对于大白菜的多态性分子标记和BoFLC3基因特异标记选择,创制附加结球甘蓝2号染色体不同区段的大白菜—结球甘蓝CSILs,同时获得含有纯合BoFLC3和无BoFLC3的背景一致的小孢子植株,研究BoFLC3在大白菜中的表达,对大白菜FLC同源基因及其他开花基因的影响,以及对抽薹开花的影响。CSILs的创建为获得带有结球甘蓝有益性状的大白菜,开展目标基因精细QTLs定位和功能基因组研究奠定基础,为芸薹属优异新种质的创建提供新途径;同时利用特色种质资源(有BoFLC3和无BoFLC3的背景一致的小孢子植株)分析BoFLC3对大白菜的影响,对深入了解FLC同源基因的作用,探索大白菜春化机制,创建耐抽薹大白菜新种质具有重要科学意义,也为研究近缘种属同源基因间的互作提供新途径

The progenies from Chinese cabbage-cabbage monosomic alien addition line added the No.2 chromosome and BoFLC3 of cabbage were used to creat Chinese cabbage-cabbage chromosome segment introgression lines (CSILs) by backcrossing, microspore culture and molecular marker-assisted selection. Chinese cabbage-cabbage CSILs with BoFLC3 and CSILs without BoFLC3 were obtained and used to study on expression of BoFLC3 in Chinese cabbage, effect on homologous genes of FLC and others genes related flowering. These CSILs could be widely used in development of Chinese cabbage added beneficial traits from cabbage, fine mapping of QTLs and functional genomics researches. And it provide new way to creat new germplasm of Brassica. Study on BoFLC3 in Chinese cabbage, which provided new way to explore genes interaction of related species and offered an opportunity to explore vernalization mechanism of Chinese cabbage and cabbage.
