以芸薹属蔬菜榨菜与紫甘蓝嫁接嵌合体有性自交后代群体为材料,通过MSAP分析群体DNA甲基化的表观遗传结构,阐明嫁接诱导表观遗传标记与获得变异表型性状的相关性以及遗传规律;通过建立有性自交第六代GS6表观自交系epiIL,与亲本榨菜杂交创建DNA序列未改变的F2表观遗传群体,分析群体的基因组甲基化状态,将多态性标记与叶形变异等性状进行单标记关联分析以及候选基因的功能验证,明确DNA甲基化变异位点与变异性状的关系及其遗传机制;通过small RNA测序和DNA甲基化测序,找到榨菜和嵌合体有性后代全基因组DNA甲基化差异区域内的24nt-siRNA表达与异源small RNA转运的关系,阐明异源small RNA调控甲基化产生的分子机理。研究结果将明确嫁接获得变异性状遗传的物质基础,阐明甲基化调控变异表型性状产生的遗传规律与分子机制,为农作物嫁接“获得性遗传”理论提供确凿的分子证据。

This project is on basis of acquiring sexual progenies with the phenotypic variation induced by grafting in Brassica (Brassica juncea and Brassica oleracea ).Firstly, in order to study the genetic regularity of epigenetic marks induced by grafting and the correlation with acquired Characters, MSAP was used to analyze the genomic DNA methylation of GS1 population(the first generation of chimera).Secondly, through analyzing the DNA methylation of epigenetic population without the alteration of DNA sequence constructed by hybridization of GS6 and Brassica juncea, polymorphic MSAP markers were linked with the alteration of leaf shape by using the single MSAP molecular marker analysis to clarify the relationship between the alteration of leaf shape and the variation loci of DNA methylation. Finally, small RNA sequencing and MeDIP-Seq were used to explore the expression of 24nt-siRNA in differentially methylated region of GS6 and Brassica juncea to illustrate the molecular mechanism that how heterologous small RNAs regulate DNA methylation. This project will clarify material basis, genetic rules and mechanism of acquired characters induced by grafting and provide a new theory of inheritance of acquired characters.
