血管新生在褥疮组织修复过程中起着至关重要的作用,它不断给新生的组织提供氧气和营养成分。目前研究发现VEGF/VEGFR-2信号通路调控着组织血管新生,该通路与艾灸促进褥疮组织血管新生密切相关。本课题基于血管新生VEGF/VEGFR-2信号通路,探讨艾灸促进褥疮血管新生的分子机制。我们拟采用激光散斑成像、Western blot、Real Time-PCR和TUNEL等技术检测褥疮组织面积、局部血流、微血管密度及其信号通路中标志性信号分子的蛋白和mRNA表达;明确艾灸促进褥疮血管新生的作用;阐明艾灸促进褥疮组织血管新生的分子机制。这将为临床艾灸促进褥疮,乃至慢性损伤组织愈合提供理论和实验依据。

Angiogenesis plays a vital role during the process of pressure ulcer healing, which provides nascent tissue with oxygen and nutrients constantly. Recent study found that VEGF/VEGFR-2 signal pathway regulates the tissue angiogenesis, which closely related to the angiogenesis of pressure ulcers treated by moxibustion. Base on VEGF/VEGFR-2 signal pathway, we plan to explore the mechanism of moxibustion promoting angiogenesis of pressure ulcers. The technology of Laser Speckle Imaging, Western blot, Real Time-PCR and TUNEL will be used to detect the area of pressure ulcer, local blood flow, micro vessel density, the protein and mRNA expression of key signal molecules in the signaling pathways. This will help clarifying the effect of promoting angiogenesis of bedsores with moxibustion; elucidating the molecular mechanism of moxibustion promoting angiogenesis of pressure ulcers. The results of this study will provide experimental and theoretical evidence for pressure ulcers treatment, and chronic wounds healing.
