蛋鸡脂肪肝出血综合征（FLHS）是一种营养代谢性疾病，主要特征为肝脏脂肪过度沉积，蛋鸡产蛋率显著下降，给蛋鸡养殖业造成严重的经济损失。然而其发病机制迄今尚不明确，所以探讨FLHS的分子病理机制对FLHS的早期诊断和防治具有重要意义。因此，本研究拟以海兰蛋鸡为试验对象，采用高能低蛋白日粮建立FLHS疾病模型；然后运用双向凝胶电泳（2D-PAGE）并结合酪氨酸磷酸化western-blot技术建立FLHS发生发展过程中FLHS患鸡和正常健康蛋鸡之间肝脏动态的蛋白质表达谱和酪氨酸磷酸化蛋白修饰谱，筛选出差异表达的蛋白质并进行质谱分析；同时应用生物信息学技术对差异表达的蛋白/磷酸化蛋白进行功能注释及网络构建，以发掘涉及禽类体内糖和脂类代谢网络途径及其代谢调节的一系列蛋白质，进而从分子水平上揭示FLHS可能涉及的病理分子路径，同时为寻找FLHS特异性诊断及治疗方法提供理论依据。

Fatty liver hemorrhagic syndrome (FLHS) is an avian nutritional and metabolic disease. FLHS is characterized by increased hepatic triacylglycerol content associated with liver hemorrhages, results in a sudden decline in egg production and causes great economic losses in poultry production. However, the molecular pathogenesis of FLHS is still unknown so far, suggesting that it's more important if there is some diagnosis biomarker. In this study, Hanlan layer hens will be employed to create disease model of FLHS by giving high energy/low protein diet. Subsequently, the profiles between whole proteins and phosphorylation proteins in the liver in the development of FLHS will be dynamically analyzed using 2-dimensional polyacrylimade gel electrophoresis (2D-PAGE) in combination with western blotting of specific anti-phosphotyrosine. Then the proteins or phosphorylation proteins detected will be annotated their biological functions by querying against the protein function database and the proteins interaction network will be constructed so as to identify a series of proteins which are related to carbohydrate metabolism, lipids metabolism and signal transduction. These results will be benficial to understand the pathogenesis of FLHS. At the same time, the findings will provide basic research data and a new rode for preventing FLHS so as to promote the poultry production.
