申请人系统阐明了结核保护性免疫的CD4 T细胞应答特征,首次报道IL-6R及调节性B细胞调节Th17应答的机制,为结核病疫苗研发提供了重要理论依据;发现多个结核病诊断分子标识,获得国家发明专利7项,转化临床应用1项,解决了我国长期没有自主知识产权的结核潜伏感染诊断试剂的问题;首次阐明宿主IL1B基因多态性及xCT在结核病发生中的作用机制,为宿主定向治疗结核病新策略的研发奠定了基础;阐明了H7N9禽流感病毒进化机制以及传播和致病的特征,为禽流感防控策略的制定提供了科学依据。研究成果得到同行高度认可,发表SCI论文50余篇,累计IF大于310,近五年被他引660余次。其中,第一或通信作者论文23篇。获得广东省科技二等奖1项,中华预防医学会二等奖2项。担任《Sci Rep》编委和多家SCI杂志审稿人。下一步拟在前期基础上,深入开展基于宿主基因定向治疗结核病的临床前研究,为耐药结核治疗提供新方案。

The applicant has focused on basic and translational research on tuberculosis and emerging infectious disease in attempt to solve the scientific question raised in clinical practice related to disease prevention, diagnosis and treatment. By taking advantage of clinical resources, he has found for the first time that suppressed Th17 T cell response, in addition to Th1 and multifunctional Th1 cell response, was associated with disease severity of tuberculosis diseases, as well as the development of active tuberculosis. He has further dissected three novel mechanisms underlying suppressed Th17 cell response in patients with tuberculosis. These included suppressing IL-6R receptor expression on CD4 T cells and inducing CD19+CD5+CD1d+ regulatory B cells expansion. Unexpectedly, he found that CD4+CD25+ regulatory T cells plays limited role in modulating Th17 response, despite it plays a critical role in inhibiting Th1 response (a publication in 2007 with 139 citation). Additionally, he has identified several biomarkers for potential clinical diagnostic application and developed one kit for diagnosis of mycobacterium tuberculosis infection. Furthermore, has found several host genes including IL22, IL6, IL1B, and xCT SNP are associated with susceptibility to tuberculosis. The mechanisms through which IL1B SNP regulates Mycobacterium tuberculosis induced IL-1beta expression and xCT participates in the immunopathogenesis have been dissected. Finally, his works in emerging infectious diseases have provided insight on the genesis, pathology and transmission of the H7N9 influenza viruses causing human infections in China 2013/2014, and found potential useful host biomarker for early diagnosis of severe cases of hand, foot, and mouth disease caused by enterovirus 71. With these achievements, he has published over 50 SCI paper with total impact factor over than 310 and more than 660 citation in the last five years. Among these publications, twenty-three were authored him as the first or corresponding author. Being an editor of the Scientific Reports, a journal of Nature Publishing Group since 2011, and reviewer for over than 10 SCI journals, he has edited and reviewed near 50 SCI paper in recent years. In further study, he will investigate the effect and mechanism of xCT-based host-directed therapy against tuberculosis.
