目前糖尿病、高脂血症、高血压等慢病的高发病率、高致残率、高死亡率成为困扰人类健康的主要问题。实现慢病防治关口前移急需找到新途径。本课题基于“体质土壤”学说,根据前期痰湿体质流行病学调查、基因水平检测结果,以及蛋白水平预实验结果,提出“痰湿体质是许多代谢性疾病的共同土壤,其引起人体代谢紊乱可能与免疫炎症进程、胰岛素抵抗及糖脂代谢相关蛋白分子表达失调有关,通过调节上述‘共同表达蛋白'可实现痰湿体质相关疾病的‘异病同治'”的假说。 运用蛋白芯片联合质谱技术,检测痰湿体质的“未病人群”、高脂血症边缘升高人群、2型糖尿病临床前期人群的外周血单个核细胞(PBMC)及血清蛋白表达,探查三者“共同表达蛋白”,并观察王琦国医大师“化痰祛湿方”对“共同表达蛋白”的调控效应及机制。本课题对于揭示中医痰湿体质异病同治的现代内涵,实现慢病防控,推动“九体医学健康计划”的实施,具有重要的理论和实践意义。

英文摘要

Nowadays, the increasing morbidity, disability rate and mortality of many chronic diseases such as diabetes, hyperlipidemia and hypertension have become major problems that threatened public health. New approaches need to be found to move forward the critical pass of the chronic diseases’ prevention. Based on the theory of body constitution soil and according to the previous epidemiological investigation, DNA test and protein levels pre-test of Phlegm-dampness constitution, this research proposed the hypothesis that the Phlegm-dampness constitution is the common basis of metabolic diseases and the mechanism that the Phlegm-dampness constitution and its related diseases can cause metabolic disorders may be related to immune and inflammatory processes, insulin resistance and the expression disorders of glucose and lipid metabolism related proteins. So the same treatment to different diseases of the Phlegm-dampness constitution related diseases can be realized by adjusting the common expression proteins mentioned above. . By using the protein chip combined with mass spectrometry technology,this research will detect the peripheral blood mononuclear cell (PBMC) and serum protein expression of Phlegm-dampness constitution individuals among pre-disease populations, hyperlipidemia populations and type II diabetes populations of preclinical stage. Then detect the common expression proteins of the three groups in order to further observe the regulating effects and mechanisms that Huatan Qushi decoction (Invented by the Master of Traditional Chinese Medicine-Wang Qi) has on the common expression proteins. . This research has important theoretical and practical significance on revealing the modern connotation of same treatment to different diseases of the Phlegm-dampness constitution, achieving the goal of better prevention and control of chronic diseases and promoting the wide application of the nine body constitution medical health plan.
