方剂是中医临床治疗疾病的药物,中药饮片是制成方剂的原料,中药有效成分应该是方剂对证治疗有效状态下的体内显效成分。类方蕴含着方剂随证衍变而表达体内显效成分的共性规律和个性差异,借助源于互联网的大数据技术能够从类方对证治疗过程中挖掘体内成分与治疗效应的关联关系。关黄柏是黄柏发挥滋肾阴降相火功效的道地药材,其药效物质基础不清影响了关黄柏的质量控制和优势疗效的发挥。本课题以关黄柏及相关滋肾降火类方(滋肾丸、大补阴丸、知柏地黄丸)为研究对象,以临床肾阴虚火旺证生物标记物为指标桥接动物模型,分析关黄柏及此类方分别在治疗肾阴虚火旺证的过程中,方剂配伍、证候体征要素、证候生物标记物、神经内分泌和免疫系统指标、中药体内成分和药动学参数等所发生的变化,利用大数据技术,挖掘类方治疗效应各指标与关黄柏体内成分的关联关系,从中发现关黄柏滋肾阴降相火功效的物质基础,为关黄柏专属性质量控制方法及创新药物设计提供依据。

Prescription is drug for disease in clinic, and Chinese herbal medicine is raw materials made for prescription, and the active ingredient in TCM prescription should be effective state components under in vivo treatment. Classified prescriptions have the common law and personality differences in vivo expression effective components. With the big data technology comes from internet, it can mine in vivo markedly relationship between therapeutic effects and effective components in the treatment process. The cortex phellodendri amurensis (Guanhuangbai) is genuine regional drug that huangbai play efficacy of nourishing ShenYin and falling XiangHuo. The unclear material basis affected the quality control and efficacy advantages of Guanhuangbai. Therefore, this project is taken Guanhuangbai and its classified prescriptions (Ziyin Wan, Dabuyin Wan, Zhibaidihuang Wan) as the research object, to investigating the changes of syndrome signs elements, syndrome biomarkers, neuroendocrine and the immune system, ingredients in vivo and pharmacokinetic parameters of Guanhuangbai and its classified prescriptions protects against ShenYin and falling XiangHuo. The use of big data technologies, it mines the connection relation between composition in vivo and pharmacokinetic parameters of Guanhuangbai and each index of therapeutic effects. Eventually clarifying the efficacy material basis of Guanhuangbai nourishing ShenYin and falling XiangHuo, it provides the basis for the quality control of Guanhuangbai and innovative drug design.
