围绝经期综合征(PMS)是影响女性健康的重要疾病,表现为神经及内分泌系统失调、植物神经功能紊乱、心理精神症状等一系列临床症状。申请者前期的研究工作表明,肾虚是围绝经期的基本生理病理特点,肝郁是PMS的核心病机,贯穿疾病全过程。为进一步阐明PMS肝郁与肾虚的机理及不同证候产生的分子生物学机制,本研究在前期基础上,采用动物实验的方法,以自然衰老和束缚法建立PMS大鼠模型,并进行以下研究:(1)分别以疏肝、补肾、疏肝补肾的代表方剂为探针,以方测证,确定PMS大鼠模型的中医证候特征;(2)以NMR手段检测并分析用药前后代谢底物的变化,研究肝郁和肾虚的NMR代谢组学特征;(3)分析PMS的细胞信号通路变化,阐明肝郁、肾虚病理、证候可能的微观机制。本项目以探索绝经期前后NMR代谢组学特征为起点,观察PMS的代谢组学相关标记物,能够进一步丰富PMS中医病因病机理论,为本病的辨证和中医药防治提供依据。

Perimenstrual syndrome (PMS) affects millions of women and it is of great importance to decipher its underlying courses for diagnosis and treatment of the disease. Our earlier work showed that the kidney deficiency (KD) was the basic pathophysiological characteristics of PMS, yet liver stagnation (LS) is the key cause through the whole course. To elucidate the syndromic basis and to analyse the PMS-associated KD and LS, a rat model for PMS will be established, so the metabolic characteristics of DK and LS can be investigated by NMR spectroscopy after the intervention by established prescriptions. The following researches are proposed: 1)investiate KD and LS’s pathophysiological appearance in rats,with the method of ‘by means of prescriptions to speculate the symptoms’; 2) characterize the metabolic changes before and after the treatment by NMR; 3) Based on the metabolic patterns, establish a link between the disease and the possible cellular signal transduction pathways. These works will allow us to set a plateform for in-depth study on differentiation of KD and LS, enriching our Chinese medicinal understanding on PMS, and providing a theoretical basis for diagnosis and treatment of the disease.
